Vanwege de verscheidenheid aan onderdelen vereist een COLTRIMS setup een vrij hoog niveau van technische expertise, met name op het gebied van vacuüm techniek, deeltje detection, snelle elektronica en data-analyse. Voordat ik overga tot het onderzoek van complexe soorten, moet het dus worden grondig gecontroleerd als de setup goed, bijvoorbeeld draait door het uitvoeren en analyseren van een meting op een diatomic of triatomisch soorten.

Chiraliteit is een functie van onze natuur die al meer dan 150 jaar fascinerende onderzoekers. In de 19e ontdekt eeuw, Pasteur, van't Hoff en anderen dat moleculen kunnen optreden in twee spiegelbeeld structuren die niet super-waardoor - zoals onze linker- en rechterhand. Deze eigenschap werd genoemd 'chiraal centrum', van het Griekse woord voor 'hand'.
Tot nu toe is geen verschil in thermodynamische eigenschappen of in de energieniveaus van links - en rechtshandig vormen (de twee ' enantiomeren') gevonden. Om de handigheid van een monster te analyseren en te scheiden van de enantiomeren, kan interactie met andere chirale moleculen worden gebruikt, zoals bijvoorbeeld in verschillende chromatographical benaderingen wordt gedaan. 1 Chiroptical methoden zoals (vibrationele) circulair dichroïsme, (V) CD en optische hiervoor dispersie, ORD, worden routinematig die onderscheid maken tussen enantiomeren. 2
Als het gaat om de bepaling van de microscopische structuur, vereisen deze technieken extra informatie, bijvoorbeeld van quantum-chemische berekeningen. De enige techniek die is algemeen aanvaard om rechtstreeks de absolute configuratie is abnormaal Röntgendiffractie. 3
Onlangs is gebleken dat de absolute configuratie van eenvoudige chirale soorten kan worden bepaald door Coulomb explosie Imaging. 4 , 5 in deze aanpak, moleculen in de gasfase zijn vermenigvuldigen geïoniseerd zodat de resterende kernen sterk elkaar afstoten. Deze afkeer leidt tot snelle fragmentatie ('explosie') van de moleculen. De richting en de omvang van het fragment momenta correlaat aan de structuur van de molecule – voor kleine molecules, de dynamiek richtingen overeen met verrassend goed de obligatie-assen. Coulomb explosie voor Moleculaire structuurbepaling heeft zijn baanbrekend werk verricht met behulp van molecuulion balken uit een versneller. 6 deze lichtbundel folie techniek heeft onlangs ook toegepast voor chirale erkenning. 7
In tegenstelling tot abnormale röntgendiffractie mag het monster geen kristallijne maar verstrekt in de gasfase. Dit maakt de Coulomb explosie aanpak ideaal voor vluchtige soorten en dus aanvulling op Röntgendiffractie. In bepaalde gevallen kan zelfs de rechts-of linkshandigheid worden bepaald voor individuele moleculen.
In de praktijk, heeft de exacte wederopbouw van de moleculaire structuur moeilijk zelfs voor methaan derivaten, bijvoorbeeld moleculen met een centrale koolstof en verschillende substituenten bewezen. Dit wordt toegeschreven aan het feit dat de interactie tussen de fragmenten niet precies Coulombic is en dat niet alle obligaties tegelijk breken. Met het oog op de stereochemische informatie, met name om te onderscheiden tussen enantiomeren, is deze reconstructie gelukkig niet nodig. In plaats daarvan kunnen de vectoren van de dynamiek van de verschillende fragmenten worden gecorreleerd zodanig dat deze een aantal dat is verschillend voor de links - en rechtshandig moleculen. Om betrouwbare resultaten, moeten ten minste vier fragment momenta worden geregistreerd.
Om te meten deze dynamiek informatie, moeten de fragmenten uit één- en slechts één-moleculaire uiteenvallen worden gedetecteerd in de stap van een enkele meting. Deze aandoening wordt meestal aangeduid als "samenvallende detectie". Bovendien, moeten de richtingen van de emissie worden geanalyseerd, die van invloed zijn bedragen in de praktijk om de tijd en de positie van het fragment te registreren in een lijst-modus-gegevensindeling.
In de atomaire en moleculaire fysica, zijn technieken ontwikkeld, die deze benadering van de meting worden geïmplementeerd door gebruik te maken van elektrostatische spectrometers voor massale scheiding en en positie-tijdgevoelige multi-hit detectoren. Het meest prominente voorbeeld is de COLTRIMS (koude Target terugslag Ion Momentum spectroscopie) setup – ook bekend als reactie Microscoop. 8 , 9 een schets voor dit soort experimenten wordt gegeven in Figuur 1. In tegenstelling tot een standaard COLTRIMS die elektronen ook kan opnemen, vereist Coulomb explosie Imaging alleen de ion detector.
Spectrometer en detector zijn gemonteerd onder ultra hoog vacuüm (&#60; 1 x 10-9 hPa) om te voorkomen dat de oprichting van ionen uit residuele gas. Afzonderlijke moleculen van het monster worden geleverd via een gasvormige gratis moleculaire straal gemaakt door supersonische uitbreiding: op grond van de dampdruk, de moleculen uit te breiden door middel van een kleine spuitmond (ongeveer 50 µm diameter) in het vacuüm. Dit deel van het experiment, de kamer van de bron, wordt gescheiden van de interactie-regio door meestal twee skimmers en differentieel gepompt stadia. Een extra differentieel gepompt sectie ligt direct achter de interactie regio te dumpen van de gas-jet en dus vermijden achtergrond gas in de regio van de interactie.
De ioniserende straling snijdt met behulp van de moleculaire straal onder 90°. De meeste laboratoria gebruiken tegenwoordig femtoseconde laserpulsen, hoewel synchrotronstraling, snelle ionen of elektron effect mogelijk 'projectielen' voor het opwekken van Coulomb explosie.
Het volgende protocol maakt de veronderstelling dat een setup uitvoeren voor samenvallende beeldvorming van ionen en een laser femtoseconde zijn beschikbaar in het lab. De piek intensiteit die nodig zijn voor het opwekken van Coulomb explosie in vier of zelfs vijf fragmenten moet worden over de volgorde van 6 x 1014 W/cm2. Om te voorkomen dat de buitengewoon lange metingen, moet de herhaling van de laser 10 kHz of meer. Dit is van cruciaal belang, want aan de ene kant, samenvallende detectie alleen kan worden vastgesteld als de kans voor versnippering in de laser focus ligt aanzienlijk lager dan 1 per laser pulse (idealiter niet meer dan 10%). Het percentage van de totale fragmentatie, aan de andere kant, mag niet lager zijn dan een paar kHz omdat het aandeel van de relevante multifragmentation trajecten meestal minder dan 10-4 is. Zo bemoedigend feit, dient te worden vermeld dat in principe al een interne versnippering gebeurtenis is voldoende om te identificeren van de configuratie van een monster enantiomeer en die detectie van een paar honderd toelaat om te bepalen van de overvloed van de enantiomeren in een steekproef van de onbekende Enantiomere samenstelling.

Dit artikel toont hoe de COLTRIMS (koude Target terugslag Ion Momentum spectroscopie) of de "reactie Microscoop" techniek kan worden gebruikt om te onderscheiden tussen enantiomeren (stereo-isomeren) van eenvoudige chirale soorten op het niveau van afzonderlijke moleculen. In deze benadering een gasvormige moleculaire straal van het monster breidt uit naar een vacuuemcel en kruist met de laserpulsen femtoseconde (fs). De hoge intensiteit van de pulsen leidt tot snel meerdere ionisatie, ontsteken een zogenaamde Coulomb explosie die verschillende kationische (positief geladen) fragmenten produceert. Een elektrostatisch veld begeleidt deze caties op tijd en positie gevoelige detectoren. Gelijkaardig aan een massaspectrometer time-of-flight, de aankomsttijd van elke ion levert informatie over de massa. Als een overschot, is het elektrostatische veld aangepast op een manier dat de richting van de emissie en de kinetische energie na fragmentatie tot variaties in de time-of-flight en in de positie van invloed op de detector leiden.
Elke ion effect creëert een elektronisch signaal in de detector; Dit signaal is behandeld door Hoogfrequent elektronica en gebeurtenis door gebeurtenis geregistreerd door een computer. De geregistreerde gegevens komen overeen met de tijden van de gevolgen en posities. Met deze gegevens, kunnen de energie en de richting van de emissie van elk fragment worden berekend. Deze waarden zijn gerelateerd aan de structurele eigenschappen van de molecule onderzochte, dat wil zeggen aan de bindingslengte en de relatieve posities van de atomen, waardoor bepalen molecuul door molecule de handigheid van eenvoudige chirale soorten en andere isomere functies.

In dit deel tonen we de resultaten voor de halomethanes. Deze soorten zijn ideaal voor het aantonen van de principiële experimenten door hun eenvoud en hoge dampdruk. In de tussentijd heeft de meer complexe soorten halothaan onderzocht met behulp van één soft-x-ray fotonen van een synchrotron bron voor het opwekken van meerdere ionisatie. 14
