אנו מציגים immunoprecipitation ששונה כרומטין מקורית של רצף (צ'יפ-seq) נסיוני תואם עם תערובת גאוסיאנית ההפצה מבוססת ניתוח מתודולוגיה (צפיפות נוקלאוזום שבב-seq; ndChIP-seq) המאפשר את הדור של מדידות משולב של נוקלאז micrococcal (MNase) נגישות עם שינוי היסטון הגנום כולו. נוקלאוזום עמדה צפיפות המקומי ואת השינוי posttranslational של subunits שלהם היסטון, לפעול בהופעה לווסת את הברית שעתוק מקומיים. מדידות קומבינטורית של נוקלאוזום נגישות עם שינוי היסטון שנוצר על ידי ndChIP-seq מאפשר החקירה בו זמנית של תכונות אלה. המתודולוגיה ndChIP-seq ישימה על מספר קטן של תאים ראשי נגיש cross-linking מבוסס שבב-seq פרוטוקולים. יחדיו, ndChIP-seq מאפשר מדידת שינוי היסטון בשילוב עם צפיפות נוקלאוזום מקומיים כדי להשיג תובנות משותפות מנגנונים המסדירים שעתוק RNA בתוך התא הראשי נדיר אוכלוסיות חדשות.
