Fire trin i protokollen er kritisk: 1) forberede HBEGF koncentration til netop 5 ng/mL, da små stigninger i koncentrationen kan føre til umodne kulturer, fx10 ng/mL HBEGF vil de differentiere astrocytter4; 2) undgå boblerne i de løsninger, som indeholder væv eller celler, som kan ændre pH og hindrer astrocyte sundhed; 3) pre-equilibrating medier til at opnå den optimale pH og temperatur til at dyrke sunde astrocytter; 4) udveksler kun halv medium en gang om ugen fordi enhver rester...

Astrocytter påvirke hjernefunktion gennem trofiske støtte, blodgennemstrømning, synaptic signalering og plasticitet og intercellulær kommunikation - alle er mekanismer, at centrum omkring de tynde astrocytic processer. AWESAM protokollen beskrevet her giver mulighed for undersøgelse af disse processer uden indblanding fra neuroner og andre glia, som er nyttige f.eks., fordi udtryk for mange proteiner og endda Ca2 + signalering overlapper hinanden på tværs af forskellige hjerne celle typer. Yderligere, denne metode overvinder begrænsningerne af tidligere tilgængelige teknikker. Især vores protokol giver i vivo-som morfologi (Stjernehus astrocytter med tynde processer) og genekspression i en hurtig, let og billig måde.
Celle morfologi, Gen-ekspression, og mange andre lovgivningsprocesser er direkte påvirket af miljøet. I en kultur parabol refererer dette til faktorer udgivet af omkringliggende celler, men også det medium, hvori cellerne er vokset. Astrocytter, HBEGF var tidligere rapporteret at fremkalde en Stjernehus morfologi1 men fandtes også at de differentiere astrocytter2. Dog lavere koncentrationer af HBEGF blev senere brugt til at generere mere i vivo-ligesom astrocytter, identificeret via RNA sequencing i to forskellige protokoller3,4. Desuden astrocyte morfologi ændres med medium sammensætning, uanset protokol4: Neurobasal medium med en lav koncentration af HBEGF er optimal for voksende Stjernehus astrocytter, mens andre mellemstore kompositioner (f.eks., serum-holdige DMEM) producerer polygonal celler (selv i astrocytter frisk isoleret af immunopanning).
AWESAM protokol overvinder ulemper af tidligere teknikker til dyrkning astrocyte monokulturer4. Tidligere teknikker til dyrkning astrocyte monokulturer har følgende ulemper: polygonal morfologi, som er ukarakteristisk Stjernehus astrocytter i vivo (MD metode)5; længde og udgifterne til protokollen (forberede astrocytter fra inducerede pluripotente stamceller tager tre måneder og kræver mange reagenser, herunder dyre vækstfaktorer)6; de lave mængder af materiale (immunopanning metode)3; og nedtrapning differentiering af astrocytter og kravet om en 3D matrix (Puschmann mfl.) 1 , 2. derimod AWESAM-protokollen giver: i vivo-indkvartering morfologi (Stjernehus astrocytter med tynde processer); en hurtig, nem, og billig metode; store mængder af materiale; og mest i vivo-indkvartering genekspression sammenlignet med immunopanned og MD astrocytter. Derudover 2D kultur giver mulighed for undersøgelse af Ca2 + signalering og vesikel genanvendelse i tynde processer og begivenheder tæt på membranen (fxJOHANNAS mikroskopi er ikke muligt i 3D kulturer).
Metoden MD har været flittigt brugt siden offentliggørelsen i 19805, tilbyder en enkel og hurtig teknik til polygonal astrocyte monokulturer. Kort sagt, MD metode indebærer voksende blandet hjerneceller i føtalt kalveserum (FCS)-indeholdende DMEM, efterfulgt af omrystning trin, berige for astrocytter (som alle andre hjerne celle typer løsnes fra fadet) og yderligere kultur i samme medium. DMEM suppleres med FCS bruges til mange andre celletyper, lige fra fibroblaster og adipocytter til forskellige kræft cellelinjer, som alle deler polygonal morfologi udstillet af MD astrocytter i kultur. Indtil den tidlige 2000s7, lille tanke havde været givet til medierne skræddersyet til astrocytter, specielt til at favorisere deres typiske Stjernehus morfologi fundet in vivo. En protokol udgivet i 2011 opnå sådanne Stjernehus morfologi: kaldte metoden immunopanning, den beskæftiger serumfrit medium optimeret til dyrkning astrocytter3. Brug denne metode, er frisk isolerede hjerneceller udsat for en række retter belagt med antistoffer, der er målrettet celle typespecifikke celle overflade proteiner, at berige for astrocytter. Trods de mere i vivo-ligesom morfologi og udtryk profil af immunopanned astrocytter, flertallet af in vitro- undersøgelser stadig stole på metoden MD astrocyte. MD-metoden er enkel og hurtig, mens immunopanning metoden består mere kompliceret og tidskrævende trin på den første dag i kultur (såsom længere enzym fordøjelsen perioder, omhyggelig lagdeling af løsninger af forskellig densitet, immunopanning, sig selv, og flere centrifugering trin - alle før plating). Men ved hjælp af mere specialiserede medium, metoden AWESAM tilbyder både hastigheden af metoden MD og i vivo-indkvartering morfologi af immunopanning-protokollen.
Samlet set AWESAM protokol er nyttigt for at studere mere i vivo-indkvartering astrocytter i 2D monokulturer isoleret fra neuroner og andre glia (som tidligere karakteriseret4 af immunostainings, immunoblots og RNA sekventering). Det giver mulighed for undersøgelse af tynde astrocytic processer, og giver stor tilgængelighed til at visualisere spontan Ca2 + signalering og begivenheder tæt på membran (fxafbildet af JOHANNAS mikroskopi).

culturing in vivo-like murine astrocytes using the fast, simple, and inexpensive awesam protocol

AWESAM (en low-koste easy stellate enstrocyte method) protokollen indebærer en hurtig, enkel og billig måde at generere store mængder af in vivo-som mus og rotter astrocyte monokulturer : Hjerne celler kan isoleres fra forskellige hjerneregioner, og efter en uge med cellekultur, ikke-astrocytic celler er rystet ved at placere kultur retter på en shaker til 6 h i rugemaskinen. De resterende astrocytter er derefter passaged til nye plader med en astrocyte-specifikke medium (betegnes NB + H). NB + H indeholder lave koncentrationer af heparin-binding EGF-lignende vækst faktor (HBEGF), som bruges i stedet for serum i medium. Efter voksende i NB + H, har AWESAM astrocytter en Stjernehus morfologi og funktion fine processer. Disse astrocytter har desuden mere i vivo-indkvartering genekspression end astrocytter genereret af tidligere udgivne metoder. Ca2 + imaging, vesikel dynamics og andre begivenheder tæt på membranen kan således studeres i fine astrocytic processer i vitro, fxved hjælp af levende celle Konfokal eller JOHANNAS mikroskopi. Især, udstille AWESAM astrocytter også spontan Ca2 + signalering ligner astrocytes i vivo.

Astrocytter, der er co kulturperler med neuroner og dyrket i NB + medium vises Stjernehus efter 2 uger af kulturen (figur 2). Ud over NB + medium, er co kulturperler astrocytter også udsat for ukendte neuron-afledte faktorer, der sandsynligvis bidrager til deres overlevelse og morfologi.
