Wondgenezing is een complex proces dat leidt tot het herstel van weefsel integriteit na letsel. Dit proces is onderverdeeld in vier overlappende stadia omsloten door hemostase, ontsteking, proliferatie en remodeling, die elk geregeld zijn door een complexe combinatie van oplosbare signalen, cel-matrix interacties en cel-cel communicatie. 1 , 2 de orkestratie van deze signalen regelt een veelheid van cellulaire reacties in de wondgenezing en bovenal cel migratie. 3 , 4 cel migratie is een zeer dynamisch proces afhankelijk van zowel cellulaire fenotypes en de biochemische en biofysische eigenschappen van de extracellulaire matrix milieu. 5 cellen voortdurend sonde hun milieu van de extracellulaire matrix door middel van integrine-gemedieerde trajecten; Dit proces maakt cellen te voelen van een breed scala van matrix eigenschappen met inbegrip van de topografie, stijfheid en opsluiting. Tweedimensionale (2D) analyse van cel migratie blijkt dat migratie wordt gedreven door lamellipodia formatie op geavanceerde kennisgebieden door middel van de generatie van actine-filament-bundels. Latere vorming van focal verklevingen op geavanceerde kennisgebieden, in combinatie met actomyosine gedreven contractie en intrekking bij de afvallende flank, rijdt de cel uit. 6 de cellulaire migratie three-dimensional (3D) op dit moment niet goed wordt begrepen, recente studies tonen echter aan dat 3D migratie is beduidend anders dan de hierboven mechanisme in 2D beschreven en waarschijnlijk door alteratief mechanismen gedreven wordt. Bijvoorbeeld, recente studies door Petrie et al. aangetoond dat, afhankelijk van de eigenschappen van de ECM, cellen in 3D-matrices tussen lamellipodium en lobopodium gedreven mechanismen van migratie schakelen kunnen. 7 omdat cel migratie een essentieel onderdeel van de wond genezing reactie is, 3D-modellen van cel migratie zijn cruciaal voor het begrip van fysiologische reacties op verschillende stimuli tijdens de wondgenezing. Hoewel cel trekgedrag op 2D oppervlakken is aangetoond dat variëren sterk van migratie in 3D-matrices, meest recente in vitro modellen van cel migratie tijdens het genezingsproces, met inbegrip van de gevestigde kras assay, wond gebruiken 2D enkelgelaagde culturen. 5 , 6 , 7 , 8 , 9 , 10 , 11 , 12 , 13 , 14 meer onlangs ontwikkelde testen hebben een 3D matrix gebruikt, maar nog steeds cultuur cellen in een enkelgelaagde op de top van de matrix, dus de beperking van de mogelijkheid om echt recapituleren de driedimensionaliteit van de cel milieu in vivo. 15
Het doel van de hier beschreven methode is om te studeren cel migratie in een fysiologisch relevante 3D-model, in plaats van op een 2D ondergrond, om een meer robuuste inzicht van migratie reacties die zijn gekoppeld aan de toegepaste prikkels. Deze methode zorgt voor de evaluatie van cel migratie binnen een 3D fibrine clot matrix in aanwezigheid van de factoren die activeren of remmen trajecten die is gekoppeld aan cel migratie. Een matrix van fibrine werd gekozen voor deze methode als gevolg van de kritische rol van fibrine in een vroeg stadium van de wondgenezing; volgende letsel, een fibrine clot wordt gevormd binnen wond omgevingen te vloeien bloedverlies en fungeert als een steiger voor de eerste cel infiltratie tijdens weefselherstel en remodeling. 2 , 16 , 17 , 18 , 19 , 20 bovendien fibrine klinisch als een chirurgische sealant wordt gebruikt en de samenstelling van de mastieken is gebonden aan cel migratie en ultimate genezing van resultaten. Een eerdere studie door Cox et al. fibroblast migratie met fibrine stolsels bestaat uit verschillende fibrine en trombine concentraties met het oog op het optimaliseren van een fibrine-sealant onderzocht. Migratie van cellen uit fibrine stolsels op kunststof gerechten werd in deze studies gekwantificeerd. 20 hier presenteren we een methode waarmee de kwantificering van de migratie van de cellen binnen de 3D fibrine-omgeving. Terwijl de specifiek in dit protocol beschreven methode fibrine gebruikt, kan dit model eenvoudig worden aangepast voor het gebruik van alternatieve matrix materialen zoals gewenst, zoals collageen of andere 3D-matrices. Daarnaast presenteren wij het gebruik van de fibroblast spheroïden in deze test omdat fibroblast migratie in het bed van de wond van het allergrootste belang voor de synthese van de extracellulaire matrix en reparatie/remodelleren van weefsel is. Echter, tijdens de reparatie proces neutrofielen zijn het eerste celtype migreren naar het bed van de wond, gevolgd door macrofagen. Fibroblasten beginnen dan te infiltreren in de omgeving van de wond ongeveer 48 uur na de eerste schade, aan het begin van de proliferatieve fase van de wond genezingsproces. 19 deze test kan eenvoudig worden aangepast om op te nemen van neutrofiele granulocyten, macrofagen, of andere celtypes te evalueren hoe wijzigingen in fibrine clot structuur van invloed zijn op migratie van deze celtypes. 21
Ter uitvoering van deze bepaling, ten eerste, een sferoïde fibroblast gevormd met behulp van een wijziging van de technieken beschreven voor cultuur van de cel van de stam. 22 de fibroblast sferoïde wordt vervolgens omgezet in een 3D fibrine-matrix, waarna uitgroei kan worden gemakkelijk gecontroleerd en gekwantificeerd gedurende meerdere dagen. Momenteel de meeste in vitro modellen van wondgenezing, zoals gevestigde kras testen, betrekken studeren cel migratie en wond sluiting op twee-dimensionale oppervlakken. De fysiologische omgeving in welke in vivo wond genezing plaatsvindt is echter driedimensionale in plaats van twee-dimensionale.
