De Fischer 344 rat is steeds een belangrijk model voor tularemie vaccin ontwikkeling3. Blootstelling aan F. tularensis via de pulmonaire route is van cruciaal belang voor het aantonen van de werkzaamheid tegen bewapende vormen van F. tularensis, omdat deze vorm van aërosolen zijn geleverd. F. tularensis (Ft) veroorzaakt de ziekte bij de mens, tularemie. Wanneer de bacteriën worden overgenomen door de pulmonaire route, leidt dit tot de tularemie, die hoge morbiditeit en mortaliteit1 heeft. F. tularensis wordt beschouwd als een agent van de biothreat als gevolg van het gevaar aërosol vormen is gekoppeld, en er is momenteel geen vaccin goedgekeurd voor menselijk gebruik in de VS Een intensieve inspanning wordt momenteel gewerkt aan het ontwikkelen van vaccins en therapeutische maatregelen tegen de tularemie, ter bescherming van de menselijke bevolking tegen het illegale gebruik van deze bacteriële biothreat.
Veel van de tularemie onderzoek heeft zich gericht op het muismodel, als gevolg van de extreme gevoeligheid van muizen tot F. tularensis infectie en de prevalentie van reagentia. Echter, muizen hebben bewezen een moeilijk model voor de ontwikkeling van een vaccin, als gevolg van de moeilijkheid van het aantonen van de werkzaamheid van het vaccin in dit model2. Onlangs, de Fischer 344 rat is ontwikkeld als een model voor tularemie vaccin ontwikkeling3. De gevoeligheid van de Fischer 344 rat naar verschillende ondersoort van F. tularensis bootst menselijke gevoeligheid4, en ratten kunnen worden beschermd tegen F. tularensis pulmonaire uitdaging door vaccinatie met een levende vaccinstam bekend om te beschermen mens5,6,7. Omdat de Fischer 344 rat voor sommige functies van F. tularensis infectie van de mens modellen, is het mogelijk een nuttig model voor de ontwikkeling van een vaccin dat tegen pulmonale F. tularensis beschermt blootstelling.
Een effectief vaccin moet beschermen van mensen tegen pulmonale blootstelling aan F. tularensis. De meest waarschijnlijke pulmonaire blootstelling van bewapende F. tularensis zou aërosol bacteriën door inademing in de longen8. Aërosol generatie van F. tularensis is echter zowel gevaarlijk en omslachtige, en vereist speciale apparatuur en insluiting. Een alternatieve route van pulmonaire blootstelling in de rat thats misschien meer aanpasbaar voor meerdere laboratoria gespecialiseerde apparatuur ontbreekt via intratracheale inenting6. Deze techniek maakt gebruik van een Laryngoscoop voor de juiste plaatsing van een katheter in de luchtpijp van een narcose rat. Plaatsing in de luchtpijp, in plaats van de slokdarm, wordt gecontroleerd door een eenvoudig apparaat dat luchtstroom uit de longen visualiseert. F. tularensis wordt vervolgens geleverd in de longen door middel van de katheter door administratie met een injectiespuit, gevolgd door de invoering van lucht in de katheter om de pulmonaire levering van de bacteriën. In tegenstelling, Jemski5 eerder gemeld dat F. tularensis geënt in Fischer 344 ratten via de intranasale route niet uit de longen tot 3 dagen na inoculatie kunnen worden gekweekt, die aangeeft dat intranasale inoculatie bij ratten doet niet leiden tot directe levering van bacteriën naar de longen.
Selecteer agent vormen van F. tularensis (F. tularensis subsp. tularensis, F. tularensis subsp. holarctica) vereisen Biosafety Level 3 (BSL3) insluiting procedures, waardoor videografie zou. F. novicida (Fn) is echter vrijgesteld van select agent status als gevolg van de avirulentie bij de gezonde mens, en kan veilig worden gebruikt onder Biosafety Level 2 (BSL2) voorwaarden9,10. Bovendien, Fn dient als basis voor levende verzwakte vaccins die tegen F. tularensis pulmonaire blootstelling beschermen kunnen wanneer geleverd via intratracheale inoculatie11,12,13. De hier gepresenteerde techniek zorgt voor de studie van infecties, die optreden via de pulmonaire route met behulp van ratten als een model voor de mens. Deze techniek kan worden uitgevoerd zonder de behoefte aan gespecialiseerde aërosol genererende apparatuur. FN werd gebruikt voor de technieken hier gefilmd.
