I den aktuelle undersøgelse præsenterer vi 3D imaging for at vurdere forholdet mellem kapillærer og individuelle voksende follikler. I vores tidligere arbejde ved hjælp af den samme protokol 9, studerede vi roller i store kar, interaktioner mellem follikler, og placeringen af follikler i intakt mus æggestokkene. KLARHED passivitet tillod os at studere mikro - og makro-vasculatures, folliculogenesis og indbyrdes forhold mellem corpora lutea og follikler samt at rekonstruere den æggestokkene a...

Hårsækken er den grundlæggende strukturelle og funktionelle enhed i æggestokkene, og dens udvikling er stærkt relateret til Vaskulaturen i æggestokken. Blodkar levere ernæring og hormoner til folliklerne og således spille en vigtig rolle i vækst og modning af folliklerne1.
En kombination af teknologier, herunder selektiv blodkar markører, transgene musemodeller og farmaceutisk udvikling, har øget vores viden om æggestokkene vaskulære netværk, angiogenese og funktionen af blodkar i folliculogenesis. Æggestokken er kendt som en aktiv orgel, fordi det bygger om forskellige væv og vaskulære netværk under folliculogenesis og ægløsning. Sådanne aktive remodeling i størrelse og struktur af fartøjer, der er nødvendig for den biologiske funktion af udvikle og rekruttere follikler.
Traditionelle histologiske og histomorphometric metoder ved hjælp af æggestokkene sektioner og immunolabeling af blodkar er begrænset til todimensionale (2D) billeder2. Med udviklingen af tre-dimensionelle (3D) genopbygning teknologier, 2D-billeder af væv skiver kan skal overlappe hinanden for at gøre en 3D struktur, men denne metode stadig har nogle begrænsninger-skæring af væv kan ødelægge mikrostrukturer, nogle dele af den væv er ofte mangler, og betydelig arbejdskraft er involveret i at gøre 3D rekonstruktioner fra billeder fås fra skiver. Hele-væv 3D imaging med Konfokal mikroskopi kan løse mange af disse begrænsninger, men disse metoder er begrænset til evaluering af angiogenese i embryonale æggestokken3. Ved hjælp af hele væv clearing metoder såsom klarhed4 kan øge den visualiseret volumen for at løse disse problemer i postnatal og voksen æggestokke, og sådanne metoder giver optisk clearance af æggestokken uden nogen strukturelle deformationer. Billeddannelse af intakt æggestokken 3D arkitektur giver en nøjagtig billeddatabase efter billede analyse software, som den Imaris softwarepakke anvendt i dette arbejde.
Ombygning af æggestokken gennem voksenalderen er en del af en dynamisk fysiologiske system, og det gør æggestokken en fremragende model for efterforskning af reguleringen af angiogenese. Desuden kan evaluering af æggestokkene blodkar rolle i patologiske betingelser for den kvindelige reproduktive system såsom polycystisk ovariesyndrom og æggestokkene kræftformer studeres gennem hele æggestokkene væv billeddannelse. Udviklingen af metoden passiv klarhed og anvendelse af avancerede billede analyse software har givet detaljerede geografiske oplysninger om relationer mellem blodkarrene og æggestokkene strukturer, såsom hårsække.

Æggestokken er det vigtigste organ i den kvindelige reproduktive system og er afgørende for produktion af kvindelige mælke og til at kontrollere det endokrine system, men komplicerede strukturelle forhold og tredimensionale (3D) Vaskulaturen arkitekturer for den æggestokken er ikke godt beskrevet. For at visualisere 3D forbindelser og arkitektur af blodkar i intakt æggestokken, er det første vigtige skridt at gøre æggestokken optisk klar. For at undgå væv svind, brugte vi hydrogel fiksering-baserede passiv klarhed (Ryd Lipid-udvekslet acrylamid-hybridiseret stiv Imaging / immunfarvning/In situ-hybridisering-kompatibelt væv Hydrogel) protokol metode til at klare en intakt æggestok . Immunfarvning, avancerede multiphoton Konfokal mikroskopi og 3D billede-rekonstruktioner blev derefter brugt til visualisering af æggestokkene fartøjerne og follikulært kapillærer. Brug denne fremgangsmåde, vi viste en signifikant positiv sammenhæng (P < 0,01) mellem længden af follikulært kapillærer og volumen af follikulært væggen.

Vi tilpasset metoden passiv klarhed i en hurtig og enkel metode til passiv æggestokken clearing samtidig bevare den follikulære og vaskulære arkitektur og opnå den højeste fluorescerende signal fra mærket markører af fartøjer og follikler. Follikulært Vaskulaturen 3D arkitektur blev fastsat ved immunfarvning for CD31, en markør i endothelial celler6.
