Dans la présente étude, nous présentons afin d’évaluer les relations entre les capillaires et les follicules en croissance individuelles d’imagerie 3D. Dans nos travaux précédents, en utilisant le même protocole n° 9, nous avons étudié les rôles des gros vaisseaux, les interactions entre les follicules et l’emplacement des follicules dans les ovaires de souris intactes. Le follicule est l’unité fondamentale structurelle et fonctionnelle de l’ovaire, et son développement est très lié à la vascularisation au sein de l’ovaire. Vaisseaux sanguins d’alimentation nutrition et hormones vers les follicules et donc jouent un rôle important dans la croissance et la maturation des follicules1.
Une combinaison de technologies, y compris les marqueurs sélective des vaisseaux sanguins, des modèles de souris transgéniques et développement pharmaceutique, ont augmenté de nos connaissances sur les réseaux vasculaires ovariennes, l’angiogenèse et la fonction des vaisseaux sanguins dans folliculogenèse. L’ovaire est connu comme un organe actif car il remodèle divers tissus et réseaux vasculaires au cours de la folliculogenèse et l’ovulation. Ce remodelage actif dans la taille et la structure des navires est requis pour la fonction biologique du développement et de recrutement des follicules.
Méthodes histologiques et histomorphométrie traditionnelles en utilisant des sections ovariennes et immunomarquage des vaisseaux sanguins se limitent à des images bidimensionnelles (2D)2. Avec le développement des technologies de la reconstruction en trois dimensions (3D), des images 2D de tranches de tissus peuvent se chevaucher pour fabriquer une structure 3D, mais cette méthode a encore quelques limitations — découpe du tissu peut détruire des microstructures, certaines parties de la manquent souvent de tissu et important travail est impliqué dans la fabrication des reconstructions 3D des images obtenues par les tranches. Imagerie 3D tissus ensemble avec la microscopie confocale peut surmonter bon nombre de ces limitations, mais ces méthodes sont limitées à l’évaluation de l’angiogenèse dans les ovaires embryonnaires3. À l’aide de tissus toute compensation des méthodes telles que clarté4 peut augmenter le volume visualisé afin de résoudre ces problèmes dans les ovaires postnatals et adultes, et ces méthodes fournissent optique clairance de l’ovaire sans aucune déformations structurelles. Imagerie de l’architecture 3D de l’ovaire intact fournit une base de données de l’image exacte pour logiciel analyse d’image, tels que le progiciel Imaris utilisé dans ce travail.
Remodelage de l’ovaire tout au long de l’âge adulte fait partie d’un système physiologique dynamique, et ce qui rend l’ovaire un excellent modèle pour les enquêtes sur la régulation de l’angiogenèse. En outre, l’évaluation du rôle des ovaires des vaisseaux sanguins dans des conditions pathologiques de l’appareil reproducteur féminin tels que le syndrome des ovaires polykystiques ou des cancers de l’ovaire peut être étudié grâce à l’imagerie des tissus ovariens ensemble. Le développement de la méthode passive de la clarté et l’utilisation du logiciel d’analyse avancées d’image ont fourni des informations spatiales détaillées sur les relations entre les vaisseaux sanguins et ovaires structures telles que les follicules.

three-dimensional reconstruction of the vascular architecture of the passive clarity-cleared mouse ovary

L’ovaire est le principal organe de l’appareil reproducteur féminin et est essentiel pour la production de gamètes femelles et pour contrôler le système endocrinien, mais les complexes relations structurelles et les architectures de système en trois dimensions (3D) vasculaire de la ovaire ne sont pas bien décrite. Afin de visualiser les connexions 3D et l’architecture des vaisseaux sanguins dans l’ovaire intacte, la première étape importante doit faire l’ovaire optiquement transparent. Pour éviter le rétrécissement des tissus, nous avons utilisé l’hydrogel clarté passive axée sur la fixation (lipides clair-échangé hybridé Acrylamide rigide imagerie / immunohistochimie/In situ hybridation-compatible avec les tissus Hydrogel) méthode pour effacer un ovaire intact du protocole . Immunomarquage, avancé la microscopie multiphoton confocale et image-reconstructions 3D servaient ensuite pour la visualisation des vaisseaux ovariens et follicules capillaires. En utilisant cette approche, nous avons montré une corrélation positive significative (P < 0,01) entre la longueur des follicules capillaires et volume de la paroi folliculaire.

Nous avons adapté la méthode passive de la clarté dans une méthode simple et rapide pour l’ovaire passive de compensation tout en préservant l’architecture folliculaire et vasculaire et obtenir le plus haut signal fluorescent de marqueurs marquées des vaisseaux et des follicules.
