I den aktuella studien presenterar vi 3D-avbildning för att utvärdera relationerna mellan kapillärer och enskilda växande folliklar. Follikeln är den grundläggande strukturella och funktionella enheten i äggstocken och dess utveckling är starkt relaterat till vaskulatur inom äggstocken. Blodkärl leverera näring och hormoner till hårsäckarna och därigenom spelar viktiga roller i tillväxt och mognad av folliklar1.
En kombination av teknik, inklusive selektiva blodkärl markörer, transgena musmodeller och läkemedelsutveckling, har ökat vår kunskap om äggstockscancer vaskulär nätverk, angiogenes och funktionen av blodkärl i folliculogenesis. Äggstockarna är känt som ett aktivt organ eftersom det remodels olika vävnader och vaskulär nätverk under folliculogenesis och ägglossning. Sådana aktiva remodeling i storlek och struktur av fartyg krävs för den biologiska funktionen av utveckla och rekrytera folliklar.
Traditionella histologiska och Histomorfometriska metoder med hjälp av äggstockscancer sektioner och immunolabeling av blodkärl är begränsade till tvådimensionella (2D) bilder2. Med utvecklingen av tredimensionella (3D) rekonstruktion teknik, 2D-bilder av vävnad skivor kan vara överlappad för att göra en 3D-strukturen, men denna metod fortfarande har vissa begränsningar — snittning av vävnad kan förstöra mikrostrukturer, vissa delar av den vävnad är ofta saknas, och betydande arbete är involverad i att göra 3D rekonstruktioner från bilder tagna från skivor. Hela-vävnad 3D imaging med konfokalmikroskopi kan övervinna många av dessa begränsningar, men dessa metoder är begränsade till utvärderingen av angiogenes i embryonala äggstock3. Använda hela vävnad clearing metoder såsom klarhet4 kan öka volymen visualiseras för att lösa dessa problem i postnatal och vuxen äggstockar, och sådana metoder ger optiska clearance av äggstocken utan någon strukturell deformationer. Avbildning av 3D arkitekturen intakt äggstockscancer ger korrekt bilddatabas för bild analys programvara, såsom programpaketet Imaris används i detta arbete.
Ombyggnad av äggstocken under hela vuxenlivet är en del av ett dynamiskt fysiologiska system, och detta gör äggstocken en utmärkt modell för utredningar av regleringen av angiogenes. three-dimensional reconstruction of the vascular architecture of the passive clarity-cleared mouse ovary

Äggstockarna är det viktigaste orgeln i det kvinnliga reproduktiva systemet och är nödvändigt för produktion av kvinnliga könsceller och för att styra det endokrina systemet, men komplexa strukturella relationer och tredimensionella (3D) vaskulatur arkitekturer av den äggstockarna är inte väl beskrivna. För att visualisera 3D anslutningar och arkitekturen av blodkärl i intakt äggstockarna, är det första viktiga steget att göra äggstocken optiskt klart. För att undvika vävnad krympning, vi använde hydrogel fixering-baserade passiva klarhet (rensa Lipid-utbyts akrylamid-hybridiserad Rigid Imaging / immunfärgning/In situ-hybridisering-kompatibel vävnad Hydrogel) protokoll metod att rensa en intakt äggstock . Immunfärgning och avancerade multiphoton konfokalmikroskopi 3D bild-rekonstruktioner användes sedan för visualisering av äggstockarna fartyg och follikulär kapillärer. Använder denna metod, vi visade en signifikant positiv korrelation (P < 0,01) mellan längden av follikulära kapillärer och volym av follikulära väggen.

Vi anpassat metoden passiva klarhet till en snabb och enkel metod för passiv äggstock clearing samtidigt bevarar den follikulära och vaskulär arkitekturen och få högsta fluorescerande signalen från märkt markörer av fartyg och folliklar. Den follikulära vaskulatur 3D arkitektur fastställdes genom immunfärgning för CD31, en markör för endotelceller6.
