Vergeleken met andere chromatografische methoden waarmee de detectie van een enkele drug soort tegelijk, vermag het huidige protocol van HPLC kwantificeren gelijktijdig drie drug compounds (DOX, MMC en DOXol) in de matrix dezelfde biologische zonder naar de nood voor wijzigen de mobiele fase. Deze voorbereiding en analyse methode is succesvol toegepast om te bepalen van de farmacokinetiek en bio-verdeling van twee nanoparticle gebaseerde drug afgiftesystemen (dat wil zeggen, Liposomale DOX en DMPLN)<sup class="x...

Chemotherapie is een primaire behandeling modaliteit voor vele vormen van kanker, maar het is vaak geassocieerd met ernstige bijwerkingen en beperkte doeltreffendheid als gevolg van resistentie tegen geneesmiddelen en andere factoren1,2,3. Ter verbetering van het resultaat van de chemotherapie zijn drug combinatie regimes toegepast in de kliniek op basis van overwegingen, zoals niet-overlappende toxicities, verschillende mechanismen voor drug actie, en niet-cross drug weerstand4,5 , 6. in klinische proeven, een betere respons van de tumor werd vaak waargenomen met behulp van gelijktijdig toegediend drug combinaties in vergelijking met een regime van sequentiële drug levering7,8. Nochtans, als gevolg van sub-optimale bio-distributie van gratis drug formulieren, gelijktijdige injectie van meerdere drugs kan veroorzaken prominente normale weefsel toxiciteit die opweegt tegen het therapeutisch effect9,10,11. Nanocarrier gebaseerde drug delivery is gebleken om de farmacokinetiek en bio-distributie van ingekapselde drugs, verbetering van de tumor-gerichte accumulatie12,13,14te wijzigen. Zoals besproken in onze recente artikelen, nanodeeltjes samen geladen met synergetische drug combinaties is gebleken dat de mogelijkheid om te verzachten de problemen door gratis drug combinaties, door hun gecontroleerde temporele en ruimtelijke co levering van meerdere geneesmiddelen om tumor weefsel, waardoor de drug van de synergetische effecten tegen kanker cellen4,15,16. Dientengevolge, zijn superieure therapeutische werking en lage toxiciteit aangetoond in beide pre-klinische en klinische studies4,17,18.
Onze vorige in vitro studies gevonden dat de combinatie van twee geneesmiddelen, Doxorubicine (DOX) en mitomycine C (MMC), een synergie-effect tegen verschillende borst kanker cellen lijnen geproduceerd en, bovendien, CO laden DOX en MMC binnen polymeer-lipide hybride nanodeeltjes (DMPLN) heeft verschillende kruisresistente bijbehorende efflux pompen (bijvoorbeeld P-glycoproteïne en borst kanker resistente eiwit)19,20,21overwonnen. In vivo, DMPLN ingeschakeld ruimtelijke-temporele co levering van DOX en MMC naar sites van de tumor en verhoogde biologische beschikbaarheid van drugs in kankercellen, zoals aangegeven door matiging van de vorming van de DOX metaboliet doxorubicinol (DOXol)22. Dientengevolge, verbeterd de DMPLN tumor cel apoptosis, tumor groeiremming en langdurige gastheer overleven in vergelijking met gratis DOX en MMC combinatie of een liposomaal DOX formulering22,23,24, 25.
Analyseren van de werkelijke hoeveelheid drugs mede geleverd door een nanocarrier is van cruciaal belang voor het ontwerpen van effectieve nanoparticle formuleringen. Veel methoden zijn ontwikkeld om te analyseren van het plasma niveau van één DOX of MMC doses met behulp van hogedrukvloeistofchromatografie (HPLC) alleen of in combinatie met de Spectrometrie van de massa (MS)26,27,28 , 29 , 30 , 31 , 32 , 33 , 34. deze methoden zijn echter vaak tijdrovend en onpraktisch voor combinatietherapie zoals een groot aantal biologische monsters apart worden voorbereid voor de analyse van meerdere drugs moet (soms met inbegrip van drug metabolieten). Naast de binding aan eiwitten sterk plasma van DOX en MMC hebben rode bloedcellen ook een grote capaciteit om te binden en het concentreren van vele geneesmiddelen35,36. Dus, plasma analyse voor DOX of MMC kan het verduisteren van werkelijke bloed drug concentraties. Het huidige werk (Figuur 1) beschrijft een eenvoudige en robuuste meerdere drug methode voor de analyse met behulp van omgekeerde fase HPLC gelijktijdig uitpakken en kwantificeren van DOX, MMC en de DOX metaboliet doxorubicinol (DOXol) van volbloed en verschillende weefsels ( bijvoorbeeld tumoren). Het is met succes toegepast om te bepalen van de farmacokinetiek en de bio-distributie van DOX en MMC en de vorming van DOXol na drug levering via gratis oplossingen of nanoparticle vormen (dat wil zeggen, DMPLN en Liposomale DOX) in een orthotopically lymfkliertest borst-tumor muismodel geïmplanteerd na intraveneuze (IV) injectie22.

Combinatie chemotherapie wordt vaak gebruikt in de kliniek voor de behandeling van kanker; echter kunnen bijbehorende nadelige effecten op de normale weefsel beperken het therapeutisch nut hebben. Nanoparticle gebaseerde drug combinatie is aangetoond dat het verminderen van de problemen van gratis drug combinatietherapie. Onze vorige studies hebben aangetoond dat de combinatie van twee geneesmiddelen, Doxorubicine (DOX) en mitomycine C (MMC), een synergie-effect tegen beide RattenUitrustingen geproduceerd en menselijke borstkanker cellen in vitro. DOX en MMC mede geladen polymeer-lipide hybride nanodeeltjes (DMPLN) overgeslagen verschillende efflux vervoerder pompen die multidrug resistentie en aangetoonde verbeterde werkzaamheid in borst tumor modellen verlenen. Vergeleken met conventionele oplossing vormen, werd deze superieure werkzaamheid van DMPLN toegeschreven aan de gesynchroniseerde farmacokinetiek van DOX en MMC en verhoogde intracellulaire drug biologische beschikbaarheid binnen de tumorcellen ingeschakeld door de nanocarrier PLN.
Om te evalueren van de farmacokinetiek en bio-distributie van samen toegediend DOX en MMC in zowel gratis oplossing en nanoparticle vormen, een eenvoudige en efficiënte multi geneesmiddelenanalyse-methode met behulp van reverse-fase hoge prestatie vloeibare chromatografie (HPLC) was ontwikkeld. In tegenstelling tot de eerder gemelde methoden die DOX of MMC afzonderlijk geanalyseerd in het plasma, is deze nieuwe HPLC-methode kunnen gelijktijdig kwantificeren DOX, MMC en een grote cardio-toxisch DOX metaboliet, doxorubicinol (DOXol), in verschillende biologische matrices ( bijvoorbeeld volbloed, borst tumor en hart). Een dubbele TL- en UV absorberend sonde 4-methylumbelliferone (4-MU) werd gebruikt als een interne standaard (I.S.) voor one-step detectie van meerdere geneesmiddelenanalyse met verschillende detectie golflengtes. Deze methode werd succesvol toegepast om te bepalen van de concentraties van DOX en MMC geleverd door zowel de nanoparticle als de oplossing benaderingen in volbloed en verschillende weefsels in een orthotopic borst tumor lymfkliertest model. De analytische methode gepresenteerd is een nuttig hulpmiddel voor pre-klinische analyse van de nanoparticle gebaseerde levering van drug combinaties.

Twee antikanker geneesmiddelen, DOX en MMC, evenals de DOX metaboliet, DOXol, werden gelijktijdig ontdekt een biologische ongehinderd onder dezelfde toegepaste gradiënt HPLC staat gebruik van 4-MU als de I.S. voor zowel de fluorescentie en UV detectoren.
