Embryonale stamceller (ESC) er pluripotent celler avledet fra preimplantation embryo, som beholde evnen til selv-fornye eller skille under bestemte forhold1. Basert på disse egenskapene, ESC har blitt et nyttig verktøy for regenerativ medisin, sykdom modellering og vev tekniske studier2.
ESC var avledet for første gang fra preimplantation mouse embryoer, stammer mus ESC (mESC) linjer med lav suksess3,4. I år forble avledning effektiviteten lave på grunn av vanskelighetene ved å opprettholde pluripotency i vitro. Dessuten tilstedeværelsen av materen celler5, som bedre avledning effektivitet, fremme kolonien vedlegg, og forbedre karyotypic stabilitet6, flere modifikasjoner av protokollen føre til en forbedring i pluripotency vedlikehold. Etter denne protokollen, over 95% av utvekst formasjon skal oppnås etter den første uken av kultur (figur 1A).
