Embryonala stamceller (ESC) är pluripotenta celler från preimplantatorisk embryon, som kvar möjligheten att antingen själv förnya eller skilja under särskilda villkor1. Baserat på dessa egenskaper, ESC har blivit ett användbart verktyg för regenerativ medicin, sjukdom modellering och tissue engineering studier2.
ESC härleddes för första gången från preimplantatorisk musembryon, med ursprung mus ESC (mESC) linjer med låg framgång3,4. För år förblev härledning effektivitet låg på grund av svårigheten att upprätthålla pluripotency in vitro. Förutom förekomsten av mataren celler5, som förbättrar härledning effektivitet, främja kolonin fastsättning och förbättra karyotypic stabilitet6, flera ändringar av protokollet bly till en förbättring av pluripotency underhåll. Den viktigaste var tillägget av leukemi hämmande faktor (LIF) till mESC odlingsmedium7,8, användningen av embryon från 129S2 tillåtande bakgrunden9 och kulturen av mESC med medium kompletteras med definierade serum, fria från potentiella differentiering faktorer10. I denna artikel skall beskriva vi ett enkelt protokoll för att effektivt härleda mESC linjer från mus blastocyster. Vi följer standardprocedurer, odla embryon från tillåtande stammar på mataren celler i härledning medium kompletteras med LIF och ett serum ersätter12. Efter detta protokoll, mESC linjer kan härledas med effektivitetsvinster som är likvärdiga med dem som erhållits i 2i medium. Trots detta kan 2i läggas för att undvika eventuella problem med pluripotency underhåll eller när du arbetar med icke-tillåtande stammar.

Tack vare dessa egenskaper är mESC ett användbart verktyg i regenerativ medicin, sjukdom modellering och vävnad ingenjörsstudier. I artikeln beskrivs ett enkelt protokoll för att få mESC linjer med hög härledning effektivitet (60-80%) av odlingsskålar blastocyster från tillåtande mus stammar på mataren celler i definierade medium kompletteras med leukemi hämmande faktor. Protokollet kan också användas för att effektivt härleda mESC linjer från icke-tillåtande mus stammar, genom enkla tillägg av en cocktail av två småmolekylär hämmare till härledning medium (2i medium). Detaljerade förfaranden för utarbetning och kultur av mataren celler, insamling och musembryon, och härledning kultur och kultur av mESC linjer finns. Detta protokoll kräver inte specialutrustning och kan utföras i ett laboratorium med grundläggande däggdjursceller kultur expertis.

Efter detta protokoll, över 95% av utväxt bildandet bör uppnås efter den första veckan av kultur (figur 1A).
