Multimodal imaging tilbyder værdifulde værktøjer til CNV patologi evaluering. Her fremlagde vi en billeddannelse protokol bestående af FA, SD-OCT og automatisk segmentering til hurtig, reproducerbare og pålidelig evaluering af CNV patologi. En pause på Bruchs membran efter laser administration blev bekræftet. Desuden brug af SD-okt på dette tidspunkt også tilladt umiddelbare visualisering af mulige intraretinal og subretinal hemorrhages, som kan forvirre fortolkning af resultaterne. Den første vellykkede forsøg på at efterligne patologi af menneskelige CNV i gnavere blev påvist næsten tre årtier siden med en kryptonfluorid i lang-Evans rotter1. Derefter blev en kryptonfluorid brugt til at bryde Bruchs membran i den mest populære mus stamme, C57BL/6J2,3,4. Succesraten for CNV induktion blev bekræftet med FA og histologiske pletter. En hurtig udvikling af noninvasive billedbehandling modaliteter, som OLT, fremmet væksten i feltet af gnavere prækliniske modeller. Evnen til at overvåge morfologiske forandringer i nethinden på flere tidspunkter i det samme øje væsentligt bidrager til reduktion af brug af dyr, og øger effektiviteten i eksperimentelle undersøgelser. Histologisk vurdering af CNV-læsioner er temmelig ligetil, og kræver mærkning af unormale kar vækst omkring stedet laser administration, image erhvervelse og areal/volumen estimation ved hjælp af et billede analyse software. I modsætning hertil, indføre i vivo billeddiagnostiske modaliteter mere komplekse analyser af CNV patologi og dens fortolkning.
Her præsenterer vi en enkel og relativt hurtig metode til grade induktion, progression og regression af CNV ved hjælp af FA, SD-OLT, og metoden automatiseret segmentering i mus laser-induceret CNV model.

in vivo multimodal imaging and analysis of mouse laser-induced choroidal neovascularization model

Laser-induceret choroidal neovascularization (CNV) er en veletableret model til at efterligne den våde form af aldersrelateret makuladegeneration (AMD). I denne protokol, vi sigter mod at guide læseren ikke blot gennem de tekniske overvejelser generere laser-induceret læsioner til at udløse neovaskulær processer, men snarere fokusere på den kraftfulde oplysninger, der kan opnås fra multimodale langsgående in vivo imaging hele opfølgningsperioden.
Laser-induceret musen CNV model blev genereret af en diode laser administration. Multimodal i vivo billeddannelse teknikker blev brugt til at overvåge CNV induktion, progression og regression. Første, spektrale domæne optisk kohærens tomografi (SD-OCT) blev udført umiddelbart efter lasering for at kontrollere en pause på Bruchs membran. Efterfølgende i vivo billeddannelse ved hjælp af fluorescein angiografi (FA) bekræftede vellykket beskadigelse af Bruchs membran fra serie billeder erhvervet på choroidal niveau. Langsgående opfølgning af CNV spredning og regression på dag 5, 10 og 14 efter den lasering blev udført ved hjælp af både SD-OCT og FA. Enkel og pålidelig klassificering af utætte CNV leasions fra FA billeder præsenteres. Automatiseret segmentering til måling af total retinal tykkelse, kombineret med manuel kaliber ansøgning om måling af retinale tykkelse på CNV websteder, tillade uvildig vurdering af tilstedeværelsen af ødem. Endelig, histologisk verificering af CNV udføres, ved hjælp af isolectin GS-IB4 farvning på choroidal flatmounts. Farvning er thresholded, og området isolectin-positive beregnes med ImageJ.
Denne protokol er især nyttig i therapeutics undersøgelser kræver høj-overførselshastighed-lignende screening af CNV patologi, da det giver mulighed for hurtig, multimodale og pålidelige klassificering af CNV patologi og retinal ødem. Høj opløsning SD-OLT kan desuden optagelsen af andre patologiske kendetegnende, såsom ophobning af subretinal eller intraretinal væske. Denne metode giver dog ikke mulighed for at automatisere CNV volumen analyse fra SD-okt billeder, som skal udføres manuelt.

En boble eller subretinal blødning umiddelbart efter lasering er ikke altid synlige. Derfor, SD-okt er særligt vigtigt at kontrollere skader af Bruchs membran. Figur 1 viser et eksempel på OCT imaging på forskellige tidspunkter efter laser administration.
