Multimodale imaging biedt waardevolle hulpmiddelen voor CNV pathologie evaluatie. Hier presenteerden we een imaging protocol bestaande uit FA, SD-OCT, en automatische segmentatie voor de snelle, reproduceerbare en betrouwbare evaluatie van CNV pathologie. Een onderbreking van de Bruch membraan na laser regering werd bevestigd. Bovendien mag het gebruik van SD-OCT in dit stadium ook directe visualisatie van mogelijke intraretinal en subretinal bloeduitstortingen, die de interpretatie van resultaten verwarren kunnen. De eerste succesvolle poging om na te bootsen de pathologie van menselijke CNV bij knaagdieren werd bijna drie decennia geleden aangetoond met een laser krypton in Long-Evans ratten1. Daarna werd een krypton laser gebruikt om breken Bruch de membraan in de meest populaire muis spanning, C57BL/6J2,3,4. Het slagingspercentage van CNV inductie werd gecontroleerd met FA en histologische vlekken. Een snelle ontwikkeling van noninvasive beeldvormende modaliteiten, zoals okt, bevorderd de groei van het gebied van knaagdier preklinische modellen. De mogelijkheid om te controleren van morfologische veranderingen in het netvlies op meerdere tijdstippen in de dezelfde ogen aanzienlijk bijdraagt aan de vermindering van dierlijke gebruik en verhoogt de efficiëntie in experimentele studies. Histologische evaluatie van CNV laesies is eerder ongecompliceerd, en vereist labeling van abnormale vasculaire groei rond de site van laser administratie en beeldacquisitie gebied/volume schatting met behulp van de software van de analyse van een afbeelding. In vivo imaging modaliteiten introduceren daarentegen meer complexe analyses van CNV pathologie en de interpretatie ervan.
Hier presenteren we een eenvoudige en relatief snelle methode om de rang inductie, progressie en regressie van CNV met behulp van FA, SD-OCT, en de automatische segmentatie methode in de muis laser-geïnduceerde CNV model.

Laser-geïnduceerde choroidal neovascularization (CNV) is een reeds lang gevestigde model na te bootsen de natte vorm van leeftijdsgebonden Macula Degeneratie (AMD). In dit protocol willen we de lezer niet alleen door de technische overwegingen voor het genereren van laser-geïnduceerde laesies om te activeren neovascular processen, maar eerder de nadruk op de krachtige informatie die kan worden verkregen vanuit de multimodale longitudinale gids in vivo imaging in de follow-up periode.
De laser-geïnduceerde muis CNV model werd gegenereerd door een diode laser rechtsbedeling. Multimodale in vivo imaging technieken werden gebruikt voor het controleren van CNV inductie, progressie en regressie. Eerste, spectrale domein optische coherentie tomografie (SD-OCT) werd uitgevoerd onmiddellijk na het laseren om te controleren of een onderbreking van de Bruch membraan. Latere in vivo imaging met fluoresceïne-angiografie (FA) bevestigd succesvolle schade van Bruch de membraan van seriële beelden verworven op choroidal niveau. Longitudinale follow-up van CNV proliferatie en regressie op dagen 5, 10 en 14 na het laseren werd uitgevoerd met behulp van zowel de SD-OCT en de FA. Eenvoudige en betrouwbare sortering van lekkende CNV leasions van FA beelden wordt gepresenteerd. Automatische segmentatie voor meting van de totale dikte van het netvlies, gecombineerd met handmatige kaliber toepassing voor meting dikte van de retinale bij CNV sites, toestaan onbevooroordeelde evaluatie van de aanwezigheid van oedeem. Ten slotte is de histologische verificatie van CNV uitgevoerd met behulp van isolectin GS-IB4 kleuring op choroidal flatmounts. De kleuring is thresholded, en het gebied isolectin-positief wordt berekend met ImageJ.
Dit protocol is vooral handig in therapeutics studies vereisen hoge-doorvoer-achtige screening van CNV pathologie aangezien het toestaat snel, multimodale en betrouwbare classificatie van CNV pathologie en retinale oedeem. Bovendien, kan hoge resolutie SD-OCT de opname van andere pathologische kenmerken, zoals de vochtophoping subretinal of intraretinal. Deze methode biedt echter geen mogelijkheid voor het automatiseren van CNV volume analyse van SD-OCT beelden, die moet handmatig worden uitgevoerd.

Een zeepbel of subretinal bloeden onmiddellijk na het laseren is niet altijd zichtbaar. SD-OCT is daarom bijzonder belangrijk om te controleren of de schade van de Bruch membraan. Figuur 1 toont een voorbeeld van OCT imaging op verschillende tijdstippen na toediening van de laser.
