Flere imaging tilbyr verdifulle verktøy for CNV patologi evaluering. Her vi presentert en tenkelig protokoll bestående av FA, SD-oktober og automatisk segmentering for rask, reproduserbare og pålitelig evaluering av CNV patologi. En pause på Bruchs membran etter at laser administrasjon ble bekreftet. I tillegg tillot bruk av SD-oktober på dette stadiet også umiddelbar visualisering av mulige intraretinal og subretinal blødninger som kan forvirre tolkning av resultatene. Det første vellykkede forsøket på å etterligne menneskelige CNV i gnagere patologi ble demonstrert nesten tre tiår siden med en krypton laser i lang-Evans rotter1. Deretter ble en krypton laser brukt til å bryte Bruchs membran i mest populære musen belastningen, C57BL/6J2,3,4. Suksessrate på CNV induksjon ble bekreftet med FA og histologiske flekker. En rask utvikling av noninvasive tenkelig modaliteter, for eksempel OKT, fostret veksten av feltet gnager prekliniske modeller. Evnen å dataskjerm morfologiske endringer i netthinnen på flere tidspunkt i samme øyet betydelig bidrar til reduksjon av dyr bruk, og øker effektiviteten i eksperimentelle studier. Histologiske evaluering av CNV lesjoner er ganske grei, og krever merking av unormal vascular vekst rundt området av laser administrasjon, bildeopptak og området/volum estimering bruker en analyseprogramvare. I kontrast introdusere i vivo tenkelig modaliteter mer kompliserte analyser av CNV patologi og dens fortolkning.
Her presenterer vi en enkel og relativt rask metode klasse induksjon, progresjon og regresjon av CNV bruker FA, SD-oktober, og automatisert segmentering metoden i mouse laser-indusert CNV modell.

in vivo multimodal imaging and analysis of mouse laser-induced choroidal neovascularization model

Laser-indusert choroidal neovascularization (CNV) er en veletablert modell å etterligne den våte formen for aldersrelatert makuladegenerasjon (AMD). I denne protokollen, vi tar sikte på å lede leseren ikke bare gjennom de tekniske betraktningene generere laser-indusert lesjoner utløse neovascular prosesser, men heller fokusere på kraftig informasjon kan fås fra intermodal langsgående i vivo imaging gjennom oppfølging periode.
Laser-indusert musen CNV modellen ble generert av en diode laser administrasjon. Flere i vivo Bildeteknikker ble brukt til å overvåke CNV induksjon, progression og regression. Først spectral domene optical coherence tomografi (SD-oktober) ble utført umiddelbart etter lasering for å bekrefte en pause på Bruchs membran. Senere i vivo bildevisning fluorescein angiography (FA) bekreftet vellykket skade av Bruchs membran fra serielle bilder kjøpt på choroidal nivå. Langsgående oppfølging av CNV spredning og regresjon på dager 5, 10 og 14 etter den lasering ble utført med både SD-oktober og FA. Enkel og pålitelig sortering av lekk CNV leasions fra FA bilder vises. Automatisert segmentering for måling av totale retinal tykkelse, kombinert med manuell kaliber program for måling av netthinnen tykkelse på CNV områder, tillate upartisk evaluering av tilstedeværelsen av ødem. Endelig utføres histologiske verifisering av CNV med isolectin GS-IB4 flekker på choroidal flatmounts. Den flekker er thresholded, og isolectin-positive området beregnes med ImageJ.
Denne protokollen er spesielt nyttig i therapeutics studier som krever høy-ytelse som screening av CNV patologi som det tillater rask, flere og pålitelig klassifisering av CNV patologi og retinal ødem. I tillegg gjør høyoppløselig SD-oktober opptak av andre patologisk kjennetegn, for eksempel opphopning av subretinal eller intraretinal væske. Denne metoden gir imidlertid ikke mulighet til å automatisere CNV volum analyse fra SD-oktober bilder, som må utføres manuelt.

En boble eller subretinal blødning umiddelbart etter lasering er ikke alltid synlige. SD-oktober er derfor spesielt viktig å kontrollere skade av Bruchs membran. Figur 1 viser et eksempel på OCT bildebehandling på ulike tidspunkt etter laser administrasjon.
