قبل نقل التبني لتحريض ATCA، كانت مثقف خلايا العقدة الليمفاوية من AQP4 معبي الببتيد الفئران في المختبر في ظروف الاستقطاب Th17 أو Th1 لمدة ثلاثة أيام. المدى استقطاب المانحين CD4+ تي الخلايا كان يؤكده سيتوكين داخل الخلايا تلطيخ (ICS) ويقاس بالتدفق الخلوي (الشكل 3). تم حقن الفئران المستفيدة ساذجة عن طريق الوريد 2 × 107 المانحة AQP4 الببتيد محددة خلايا تي. بعد ستة أيام تقريبا، حوالي 100% فئران المستفيدة وضع علامات سريرية لأمراض المناعة الذاتية والملاحة والمراقبة، بما في ذلك الشلل الذيل واطرافهم هند يعرج (الشكل 4). الخلايا T AQP4 حدة الاستقطاب Th17 الناجمين عن أشد الأمراض السريرية من خلايا تي AQP4 على حدة الاستقطاب Th1. ويبين 1 فيديوماوس تمثيلية التي تلقت Th17 AQP4-الببتيد-محددة ووضع الشلل الكامل هند أطرافهم (الشلل النصفي). على النقيض من الفئران التي وضعت EAE بعد إقامة خلايا Th17 موج على حدة، استعادت الفئران المستفيدة من الأمراض السريرية الناجمة عن خلايا Th17 AQP4 على حدة. أما ع الناجمة عن خلايا Th17 p35-55-خاصة بموج، الأمراض السريرية الناجمة عن AQP4 p135-153-محددة أو خلايا Th17 p201-220-محددة مرتبطة بتسلل خلايا وحيدات النوى في الجهاز العصبي المركزي حمة والسحايا (الشكل 5). وكانت الآفات أكثر وفرة في السحايا مما في حمة للجهاز العصبي المركزي المناعة الذاتية الناجمة عن خلايا Th17 AQP4 على حدة.
مشاركة العصب البصري تجلى التقييم النسيجي، وقبل أكتوبر المسلسل AQP4-محددة وخلايا Th17 موج على حدة كلا التهاب العصب البصري المتعمد، التي تميزت بوجود خلايا وحيدات النوى. بينما تسبب خلايا Th17 الخاصة AQP4 بيرينيوريتيس الألياف الضوئية، خلايا Th17 الخاصة بموج الناجم عن التهاب العصب البصري الحاد (الشكل 5). استخدام OCT المسلسل، التهاب العصب البصري وكان واضحا من تورم وزيادة سمك الطبقة الشبكية الداخلية (أيرلندا) للأمراض السريرية الناجمة عن AQP4 محددة أو خلايا Th17 الخاصة بموج (الشكل 6). بالنسبة للجهاز العصبي المركزي التي يسببها AQP4 Th17 المناعة الذاتية، سمك IRL عاد إلى الأساس كالفئران استردادها من الأمراض السريرية. وفي المقابل، استمرار ع فعل Th17 الخاصة بموج تقابل مع أيرلندا رقيق، وكما أثبتنا سابقا17، ارتبط بفقدان خلايا الشبكية العقدة.
