La retina, EPR y coroides son tejidos complejos que muestran importantes diferencias regionales en la expresión de proteínas, función fisiológica y patológica susceptibilidad1,2. Por ejemplo, enfermedades como la degeneración relacionada de Macular (AMD), retinitis pigmentosa y la retinopatía serosa central demuestran características localización dentro de la fóvea, mácula o retina periferia1,3, 4,6. Aquí, presentamos un método de demostración de retina cómo distinta regiones pueden ser muestreadas independientemente. El objetivo general de este método es proporcionar a una guía confiable para la recolección de muestras de tejido de las regiones periféricas, maculares y foveal de la retina humana y la RPE-coroides para análisis proteómico. El fundamento para el desarrollo y uso de esta técnica es a través de análisis proteómico de estas regiones retinianas específicas, importantes penetraciones moleculares pueden obtenerse en las funciones fisiológicas y fisiopatológicas de estas regiones.
Este enfoque promete revelar la base proteómicos para susceptibilidad de enfermedad regional relativa y para facilitar la identificación de nuevas dianas terapéuticas específicas. De hecho, las investigaciones proteómicas de vítreo y sus interacciones con la retina han proporcionado ideas claves en la composición molecular y función del tejido sano y enfermo5,7,8 , 9 , 10 , 11 , 12 , 13. sin embargo, se carecen de análisis proteómicos claro comparativo de distintas regiones retinianas. La técnica ayuda a apoyar estos estudios necesaria, ofreciendo ventajas sobre otros métodos demostrando un enfoque de la colección de tejido fiable y reproducible. Más aún, el enfoque es muy accesible, tomando ventaja de sacador de tejido de tamaño estándar y disponible herramientas de biopsia. Nuestra técnica hace hincapié en la adecuada recolección y almacenamiento de tejidos para proteómica de procesamiento, haciendo consideraciones importantes para la estabilidad de la proteína y la degradación. Así, este método es más apropiado para investigadores teniendo en cuenta posterior análisis molecular de factores proteómicos.

La retina humana se compone de la neuroretina sensorial y el epitelio pigmentado retiniano subyacente (RPE), que es firmemente complexed a la capa vascular de la coroide. Diferentes regiones de la retina son anatómicamente y molecularmente distintas, facilitando funciones únicas y demostrar susceptibilidad diferencial a la enfermedad.
