Dus, dit protocol heeft tot doel te beoordelen van de mitochondriale functie door het kwantificeren van de mitochondriale Ca2 + retentie capaciteit en Ca2 +-trigged mitochondriale zwelling.
CA2 + retentie capaciteit is de meting van het vermogen van de mitochondriën om opname cytosolische calcium. Mitochondriën kunnen buffer het cytosolische vrij calcium en calcium-afhankelijke cellulaire processen regelen door opname van de calcium in de vorm van inactieve precipitaten. Verminderde Ca2 + retentie capaciteit treedt op in stress verschijnselen die is gekoppeld aan de beperking van de energie en zelfs neurodegeneratieve ziekten4,5. Geïsoleerde mitochondriën of digitonin-permeabel cellen kunnen worden gebruikt voor identificatie van de Ca2 + retentie capaciteit, en de verhoogde capaciteit om te accumuleren Ca2 + door geïsoleerde mitochondriën met de additonal glutamaat en malate wordt niet verricht door de mitochondriale isolatie procedure6. Een bepaalde hoeveelheid digitonin moet worden gebruikt om de permeabilize van de plasma-membranen van verschillende celtypes.
