Capacité de rétention en ca2 + est la mesure de la capacité des mitochondries de calcium cytosolique de l’absorption. Mitochondries peuvent mettre en mémoire tampon le calcium libre cytosolique et régulent les processus cellulaires dépendant du calcium par l’absorption de calcium sous forme de précipités inactives. Capacité altérée de Ca2 + rétention se produit dans les phénomènes de stress associés à la limitation de l’énergie et de4,maladies neurodégénératives voire5. Les mitochondries isolées ou cellules perméabilisées digitonine peuvent être utilisés pour identifier la capacité de rétention Ca2 + , et la capacité élevée d’accumuler Ca2 + par les mitochondries isolées avec le glutamate supplementaires et le malate n’est pas effectuée par le procédure d’isolement mitochondrial6. Une quantité titrée de digitonine devrait être utilisée pour permeabilize les membranes plasmiques des différents types de cellules. Le sel de hexapotassium de Ca2 +-liaison colorant fluorescent vert, un Ca2 +-cellule-imperméants sensible indicateur visible de la lumière-excitables, a été largement utilisé7. Ca2 +-vert colorant fluorescent est un indicateur de faible affinité et utilisée pour montrer que des concentrations2 + Ca libre intracellulaires continuent d’augmenter au cours de reliure prolongée de stimulations (5 min)8. Après accumulation de Ca2 + , les mitochondries dépolarisé, libéré Ca2 + dans le milieu et a commencé à enfler. La microscopie électronique et la diminution de lumière absorbance à 540 nm peut être utilisé pour mesurer le Ca2 +-déclenchée mitochondrial gonflement10,11.
