Grâce à son efficace et moins fastidieux processus d’expérimentation, technologie exempte d’étiquette sur les cellules a connu une croissance rapide ces dix dernières années à des fins analytiques ainsi que la projection comme dans l’aspect de la protéomique1,2 , livraison3, etc.4,5 comparé avec des méthodes biochimiques traditionnelles visant à l’analyse cellulaire, analyse de cellules libres étiquette en temps réel avec le prototype développé par Giaever et collaborateurs précédemment6 de drogue est basé sur le principe de l’enregistrement des modifications du signal électrique sur la surface de la cellule-attachée puces électroniques, qui permettent une mesure continue de la croissance cellulaire ou de migration de manière quantitative. Suite à cette stratégie, une cellule en temps réel (RT-CES) système utilisant le principe de détection impédance électrique était introduite7,8 et, plus récemment, un analyseur commercial cellule en temps réel (RTCA) de détection électronique a été lancée laboratoire de recherche9.
L’analyseur de la cellule en temps réel commercial principalement lit les signaux évoqués LiPo de l’impédance électrique, entraînant des changements physiologiques des cellules couvés, y compris la prolifération cellulaire, migration, viabilité, la morphologie et l’adhérence sur le surface de micropuces10,11. Ces signaux électriques est également convertis par l’analyseur dans un paramètre sans dimension nommé la cellule Index (CI) pour évaluer l’état de la cellule. Le changement de l’impédance des micropuces reflète principalement l’environnement ionique des cellules couvertes à l’interface électrode/solution. Par conséquent, les performances analytiques de l’analyseur de cellules s’appuie fortement sur l’unité de détection de base, les micropuces jetables(p. ex., ce qu’on appelle plaques électroniques, par exemple, 96/16/8 puits), qui sont faites de microélectrodes or rangés lithographically imprimé au bas du puits de l’incubation. Des microélectrodes or assemblent sous forme de cercle en ligne (Figure 1) et couvrent la majeure partie de la surface des puits d’incubation, qui permettent une détection dynamique et sensible de cellules attachées3,12,13 ,,14. Le CI augmentera dans le cas de couverture plus de la surface des cellules sur la puce et diminue lorsque les cellules sont exposées à un substance toxique, aboutissant à l’apoptose. Bien que l’analyseur en temps réel cellulaire a été fréquemment utilisé pour déterminer la cytotoxicité11 et neurotoxicité15 et fournir plus d’informations cinétiques que la méthode classique de points de terminaison, les plaques électroniques jetables sont les plus coûteuses consommable.
Jusqu'à présent, il n’y a eu aucune méthodes disponibles pour la régénération de la plaque électronique, qui est probablement dû au fait que des conditions de régénération dure, comme solution de piranha ou acide acétique sont impliqués16,17, 18, qui peuvent altérer l’État électrique de puces or. Par conséquent, une méthode douce et efficace pour enlever les cellules adhérentes et autres substances de la surface des puces or sera souhaitable pour le processus de régénération de la plaque électronique. Nous avons récemment mis au point un protocole visant à la régénération des plaques électroniques jetables, utilisant des réactifs non corrosifs et les puces régénérées sont caractérisés par des méthodes électrochimiques ainsi qu’optique19. En utilisant des réactifs de laboratoire facilement disponible et modérée comme la trypsine et éthanol, nous avons établi une méthode générale pour régénérer la plaquette commerciale électronique sans entraîner d’effets indésirables, qui est appliquée avec succès pour régénérer les deux principaux types de la plaque électronique (16 et L8) utilisé pour RTCA.

En raison de sa capacité à fournir des informations plus dynamiques sur les cellules dans des conditions physiologiques que les tests biochimiques classiques, cette analyse de cellules exempt d’étiquette en temps réel basée sur le principe de l’impédance électrique est d’attirer plus d’attention au cours des dernières décennie. Toutefois, son utilisation pratique peut être limitée en raison du coût relativement coûteux de la mesure, où coûteux consommables jetables or micropuces sont utilisés pour l’analyseur de la cellule. Dans ce protocole, nous avons développé une stratégie générale pour régénérer déployèrent microélectrodes or équipés d’un analyseur de commercial cellule exempte d’étiquette. Le processus de régénération comprend la digestion trypsique, lavage à l’éthanol et l’eau et une étape de filature. La méthode proposée a été testée et démontrée son efficacité pour la régénération et l’utilisation répétée des plaques électroniques commerciaux au moins trois fois, qui aidera les chercheurs enregistrer sur la cherté en cours d’exécution des essais sur des cellules en temps réel.
