L’hydrogel présentée dans le présent protocole (Figure 1) comporte deux grands volets : le chitosan de glycol polymère naturel et un benzaldéhyde synthétique fin gelator polymère DF PEG, qui sont les deux matériaux biocompatibles. Synthèse de DF PEG est présentée à l’aide d’une réaction de modification en une seule étape. Les hydrogels sont des matériaux polymères réticulés avec de grandes quantités d’eau et des propriétés mécaniques douces et ils ont été utilisés dans nombreuses applications bio-médicales1,2. Hydrogels peuvent offrir un environnement doux et humide, ce qui est très similaire à l’environnement physiologique des cellules in vivo. Par conséquent, hydrogels sont devenus l’un des plus populaires utilisés pour 3D cell culture3,4. Par rapport à la culture cellulaire 2D Pétri, culture cellulaire 3D a progressé rapidement pour offrir qu'une matrice extracellulaire (ECM) imité le micro-environnement des cellules à contacter et assembler pour la prolifération et la différenciation des fins5. En outre, hydrogels contenant des polymères naturels pourrait offrir des environnements biocompatibles et promotion pour les cellules à proliférer et se différencier3. Hydrogels dérivés de polymères synthétiques sont préférés pour leurs composants simples et claires, qui excluent des influences complexes comme les protéines d’origine animale ou des virus. Parmi tous les candidats d’hydrogel pour culture cellulaire 3D, hydrogels qui sont facilement préparés et ont une propriété compatible sont toujours préférés. La possibilité d’ajuster les propriétés de l’hydrogel pour s’adapter aux exigences différentes recherches est importante en tant que puits6.
Ici, nous introduisons une préparation facile d’un glycol axée sur le chitosan hydrogel utilisant la chimie dynamique imine, qui devient une plateforme polyvalente hydrogel pour 3D cell culture7. Dans cette méthode, bien connu biocompatible glycol chitosan servent à établir les cadres des réseaux de l’hydrogel. Ses groupes aminés sont réagit avec un benzaldéhyde résilié polyéthylène glycol comme le gelator de polymère forment des liaisons dynamiques imine comme crosslinkages des hydrogels8. Liaisons dynamiques imine peuvent former et se décomposent réversiblement et adaptée à un environnement, dotant les hydrogels avec réglage mécanique réticulé réseaux9,10,11. En raison de sa teneur en eau élevée, matériaux biocompatibles et résistances mécaniques réglables, l’hydrogel est appliquée avec succès comme un échafaudage pour les cellules L929 3D cell culture12,13. Le protocole ici en détail les procédures, y compris la synthèse des polymères gelator, hydrogel préparation, cellule intégration et mise en culture cellulaire 3D.
L’hydrogel montre également plusieurs autres fonctionnalités en raison de son crosslinkages imine dynamique, y compris ses multiples adaptés à divers bio-stimulants (acide/pH, pyridoxal dérivés de vitamine B6, papaïne protéine, etc.), ce qui indique que l’hydrogel peut être induit à se décomposer sous des conditions physiologiques8. L’hydrogel est également automatique guérissables et injectable, qui signifie l’hydrogel pourrait être administré via une méthode d’injection invasive minimale et obtenir un avantage dans la drogue et la cellule livraisons14,15. En ajoutant des additifs fonctionnels ou gelators polymère préconçu spécifique, l’hydrogel est compatible à l’obtention de propriétés spécifiques comme magnétique, température, pH réactif, etc.16,17, ce qui pourrait remplir un large gamme de besoins de recherche. Ces propriétés révèlent capacité potentielle de l’hydrogel pour être un injectable plusieurs porteuses des médicaments et des cellules dans des applications et de recherches bio-médicales in vitro et in vivo .

Le protocole présente une méthode facile, efficace et polyvalente pour préparer les hydrogels axée sur le chitosan aide chimie dynamique imine. L’hydrogel est préparé en mélangeant des solutions de chitosan de glycol avec un gelator de polymère synthétisé benzaldéhyde résilié et hydrogels sont efficacement obtenus en quelques minutes à température ambiante. Par divers ratios entre chitosane de glycol, gelator de polymère et des teneurs en eau, hydrogels polyvalents avec temps de gélification différents et rigidité sont obtenus. Si endommagé, l’hydrogel peut récupérer ses apparitions et du module, en raison de la réversibilité des liaisons dynamiques imine comme crosslinkages. Cette propriété se guérissables permet l’hydrogel à être injectable car il peut être autonome guéri de morceaux pressés d’un hydrogel intégrale en vrac après le processus d’injection. L’hydrogel est également multi-sensible aux nombreux stimuli bio-actifs en raison de statuts différents de l’équilibration des liaisons dynamiques imine. Cet hydrogel a été confirmé comme bio-compatibles et cellules de fibroblaste de souris L929 étaient embarqués suivant les procédures standards et la prolifération cellulaire a été évaluée facilement par un procédé de culture cellulaire 3D. L’hydrogel peut offrir une plate-forme réglable pour différentes recherches où un imitateur physiologique d’un environnement 3D pour les cellules est profité. Ainsi que ses propriétés multiples adaptées, Self guérissables et injectables, les hydrogels peut potentiellement être appliqués comme plusieurs transporteurs des médicaments et des cellules dans les futures applications bio-médicales.
