Denne protokollen er ukomplisert, er det noen viktige trinn som må utføres nøye. Flere protokoller finnes for å vurdere cytolytic (CTL) potensialet i en CD8+ eller CD4+ T-celle. Denne vurderingen kan lett gjort i vitro under kontrollerte forhold1,2,3. I tillegg infeksjonssykdommer modeller, for eksempel LCMV, klassisk har undersøkt CTL-funksjonen ved hjelp av ulikt CFSE (5-(and 6)-Carboxyfluorescein diacetate succinimidyl ester) merket målcellene hvor den CFSEHei-merkede celler pulserende peptid og CFSElo-merket målet celler igjen unpulsed. Cellene deretter injisert på en 1:1 ratio og vurdert for tap av den CFSEHei-merket pulserende mål av flyt cytometri4. Vaksine og avvisning modeller har også brukt lignende strategier for vurdering av i vivo drepe av både CD8+ og CD4+ T celler samt NK celler5,6. Dette er en kraftig analysen, men krever bruk av smittestoffer som prime immunsystemet før målet injeksjon.
Denne protokollen, derimot, krever ingen tidligere smitte av verten og utnytter stedet en vaksinasjon strategi for å prime immunsystemet før målet injeksjon. Denne vaksinasjon består av en vannbasert formulering av peptid vaksine som krever levering av en immunostimulatory cocktail kalt covax7, bestående av en Toll-like reseptor 7 (TLR7) Agonistiske (imiquimod krem), en agonistic anti-CD40 antistoff, og interleukin-2 (IL-2) fører til synergistisk kombinasjon av immunostimulatory agenter for elicitation av peptid-spesifikke grunning og robust immunrespons. Som gir denne analysen en rask avlesning CTL-funksjonen som vaksinen administreres med cellene for vurdering av funksjonen bare tre dager før injeksjon av målcellene. I tillegg er covax grunning sterk nok til at drapet kapasitet primet antigen-spesifikke T cellen kan sees mellom 4 og 24 h etter injeksjon.
Stor begrensning av denne protokollen er å identifisere den tidslinje i vivo for deteksjon av mål drapet som passer både antigen og hypotesen blir testet. Forsiktig vurdering må utføres, som variasjoner i antigen styrke samt genetiske endringer blir testet i T-celler kan føre differensial CTL-funksjon som krever forskjellige timing gjenkjenning av målet drapet. Dessuten, mens den aktuelle konsentrasjonen av peptid for vaksinasjon er optimalisert for menneskelig melanom antigen glycopeptide 100 (hgp10025-33) og ovalbumin257-264 (OVA257-264)8,9 , bruk av en annen antigen modell som kan være mer passende til en gitt studie vil kreve ytterligere validering. in vivo assay for detection of antigen-specific t-cell cytolytic function using a vaccination model

Gjeldende metoder for antigen-spesifikke drap er begrenset til i vitro bruk eller benyttet i infeksjonssykdommer modeller. Det er imidlertid ikke en protokoll spesielt ment å måle antigen-spesifikke drepe uten en infeksjon. Denne protokollen er utformet og beskriver metoder for å overvinne disse begrensningene ved å tillate for påvisning av antigen-spesifikke drepe for en Målcelle ved CD8+ T celler i vivo. Dette oppnås ved å flette en vaksinasjon modell med en tradisjonell CFSE-merket målet drepe analysen. Denne kombinasjon gjør forskeren å vurdere antigen-spesifikke CTL potensialet raskt og direkte som analysen ikke er avhengig av tumor vekst eller infeksjon.
