Under dialyse af RSF løsning er pH-værdi afgørende for følgende koncentration proces. Hvis pH-værdien afioniseret vand er mindre end 6, vil RSF løsning være lettere at gel under koncentration proces. For at undgå gellation, føjes CaCl2 til RSF løsning. Koncentrationen af CaCl2 er 1 mmol pr. vægt af RSF.

Spider og avlen kan producere fremragende silke fibre fra vandige protein løsning ved omgivende temperatur og tryk. Klipning og ekstensionelle flow kan inducere dannelse af flydende krystal tekstur i silke kirtel1. I de seneste år har der været en stor interesse i efterligne udspinding af edderkop for at producere høj styrke kunstige fibre. Dog, store mængder af spider silke protein ikke kan fremlægges, effektivt og økonomisk af landbrug edderkopper på grund af kannibalisme. Betydelige mængder af silkeormslarver silke kan opnås nemt ved landbrug. Ellers, avlen og spider har en lignende spinning proces og aminosyre sammensætning. Derfor er silkworm silke fibroin valgt som en erstatning til spin kunstige animalske silke af mange forskere.
Spider og avlen presse protein løsning gennem deres spinning kanalen i fiber i luften. Høj stress kræfter genereret langs spinning kanalen sandsynligvis stretch silke fibroin molekyler til en mere udvidet kropsbygning2. Kunstig silke fibre har været spundet ved hjælp af konventionelle våde spinning og tør-spinning processer3,4, som ikke tager hensyn de flydende styrker genereret i spinning kanalen.
Første, mikrofluid tilgange blev brugt til at undersøge forsamlingen af silke protein5,6. Derefter, mikrofluid fabrikation af RSF blev undersøgt via modellering af klipning og ekstensionelle styrker7,8. Young's modulus og diameter på RSF fibre kan indstilles af mikrofluid våde spinning, men trukket fiber trækstyrke var mindre end 100 MPa7. Endelig, højstyrke RSF fibre blev med held udarbejdet ved hjælp af metoden mikrofluid tør-spinning, men diameter af fiber er kun 2 µm8. For nylig, mikrofluid våde spinning brugt i produktionen af højstyrke rekombinant spider silke fibre. Efter spinning tegning i luften forbedret overflade og indre fejl af kunstige fibre9.
I denne undersøgelse, er de forbedrede mikrofluid spinning proces til RSF fiber indført. Det sigter mod at efterligne udspinding af silkeormslarver silke, herunder spinning dope, klipning styrker og tør-spinning proces. Denne spinning metode ikke kun kan producere høj styrke kunstig silke fiber, men også kan justere diameter af fiber. For det første var RSF spinning dope forskydes og aflange i en biomimic kanal med en anden rækkefølge eksponentiel henfald. For det andet blev påvirkninger af relativ luftfugtighed (RH) på fiber morfologi og egenskaber undersøgt i mikrofluid tør-spinning proces10. Sammenlignet med konventionel snurrende spindedyse, vores mikrofluid systemet er stærkt biomimetiske og kan bruges til at producere høj styrke fiber fra løsninger ved omgivende temperatur af tør eller våd spinning metode.
På grund af den høje opløsning, høj lysstyrke og højenergi af synkrotron stråling microfocus X-ray, kan det bruges til at karakterisere mikrostruktur af en enkelt fiber med en diameter på flere mikrometer4,11 , 12 , 13 , 14. her, SR-WAXD teknikken blev brugt til at beregne crystallinity, crystallite størrelse og krystallinsk orientering af RSF fibre.

Protokollen viser en metode til at efterligne udspinding af avlen. I den oprindelige udspinding giver ordregivende spinning kanalen silke proteiner at være kompakt og bestilt af klipning og brudforlængelse styrker. Her, blev en biomimetiske mikrofluid kanal designet til at efterligne spinning kanalen af silkworm specifikke geometri. Diffraktion buer tæt på ækvator af to-dimensionelle WAXD mønster viser, efterbehandlet RSF fiber lidt højt orientering.

Høj styrke RSF fibre blev med succes produceret ved hjælp af mikrofluid spinning metode. Stress-strain kurver og SEM billeder af strakte RSF fibre C44R40 er vist i figur 2.
