Durante la diálisis de la solución RSF, el valor del pH es fundamental para el proceso de concentración. Si el valor del pH del agua desionizado es menor que 6, la solución RSF será más fácil durante el proceso de concentración del gel. Para evitar la gelificación, CaCl2 se agrega a la solución RSF. Araña y el gusano de seda pueden producir fibra de seda pendiente de solución acuosa de proteína a temperatura y presión. Flujo extensional y corte puede inducir la formación de la textura de cristal líquido en la glándula de seda1. En los últimos años ha habido un gran interés en imitar el proceso de hilado de la araña para producir fibras artificiales de alta resistencia. Sin embargo, grandes cantidades de proteína de seda de araña pueden producir eficientemente y económicamente por agricultura arañas debido al canibalismo. Grandes cantidades de seda de gusano de seda pueden obtenerse fácilmente por cultivo. De lo contrario, el gusano de seda y la araña tienen una composición similar de proceso y del aminoácido de spinning. Por lo tanto, fibroína de seda de gusano de seda es seleccionada como un sustituto a hilar seda animal artificial por muchos investigadores.
Araña y el gusano de seda sacan solución proteica a través de su conducto de spinning en fibra en el aire. Las fuerzas de alta tensión generadas a lo largo del conducto de spinning más probable estiran las moléculas de fibroína de seda para una conformación más extendida de2. Seda artificial fibras han sido hiladas mediante hilatura convencional de húmedo y seco-spinning procesos3,4, que no toman en cuenta las fuerzas fluidas generadas en el conducto de spinning.
En primer lugar, se utilizaron enfoques de microfluidos para investigar el conjunto de proteínas de seda5,6. Entonces, microfluidos fabricación de RSF se estudió mediante el corte y extensional fuerzas7,8de modelado. Módulo de Young y diámetro de las fibras RSF pueden ajustarse por hilatura húmeda microfluídicos, pero resistencia a la tracción de la fibra dibujada era menos de 100 MPa7. Por último, fibras de alta resistencia RSF con éxito fueron preparadas utilizando el método de microfluidos seco-spinning, pero el diámetro de la fibra es sólo de 2 μm8. Recientemente, hilado húmedo microfluídicos fue utilizado con éxito en la producción de fibra de seda de araña recombinante de alta resistencia. En este estudio, se introduce la microfluídica mejorada spinning proceso de fibra RSF. Su objetivo es imitar el proceso de hilado de seda de gusano de seda, como la droga de giro, fuerzas y proceso seco-que hace girar de la corte. Este método de spinning no sólo puede producir fibra de seda artificial de alta resistencia, pero también puede ajustar el diámetro de la fibra. En primer lugar, el RSF spinning droga fue cortado y alargado en un canal biomimic con un decaimiento exponencial de segundo orden. En segundo lugar, las influencias de la humedad relativa (HR) sobre la morfología de la fibra y propiedades fueron estudiadas en la microfluídica seco-spinning proceso10. En comparación con la hilera de hilatura convencional, nuestro sistema de microfluidos es altamente biomiméticos y puede ser utilizado para producir fibra de alta resistencia de las soluciones a temperatura ambiente por el seco o mojado método de giro.
Debido a la alta resolución, alto brillo y alta energía de la radiación del sincrotrón microfocus rayos x, puede ser utilizado para caracterizar la microestructura de una sola fibra con un diámetro de varios micrómetros4,11 , 12 , 13 , 14. aquí, SR WAXD técnica fue utilizada para calcular la cristalinidad, tamaño del cristalito y orientación cristalina de fibras RSF.

El protocolo muestra un método para imitar el proceso de hilado del gusano de seda. En el proceso de hilado nativo, el contratante conducto de spinning permite las proteínas seda compacto y ordenado por las fuerzas de cizallamiento y el alargamiento.
