Veiligheidsnormen en ook studies van stamcel therapie zijn dringend nodig voordat we regeneratieve stamcel behandeling voor KOA van de Bank aan het bed brengen kunnen. Knie artrose (KOA) is een degeneratieve aandoening als gevolg van verlies van de articulaire kraakbeen en progressieve ontsteking, die uitgegroeid een grote chronische ziekte bij ouderen rond de wereld1 tot is. Huidige therapieën met anti-inflammatoire geneesmiddelen, fysieke supplementen en chirurgische procedures kunnen echter alleen tijdelijke verlichting voor symptomatische pijn2.
Menselijke adipeus afkomstige mesenchymale stamcellen (haMSCs) geworden een veelbelovende regeneratieve remedie tegen artrose van de knie, als gevolg van hun multipotente differentiatie mogelijkheden voor regeneratie van het kraakbeen en immunomodulerende eigenschappen3, 4. Vergeleken met farmacologische routes naar mechanismen van actie in vivoonderzoeken, is het bijhouden van levende haMSCs in kleine KOA diermodellen momenteel leerzaam om vast te stellen van de redenen voor en de haalbaarheid van haMSC therapie voorafgaand aan klinische toepassing. Voor preklinische testen, destabiliseert mediale meniscectomy (MM) de mechanische belasting van het gewricht te bewegen KOA in ratten, waarmee een relatief haalbaar model met consistente reproduceerbaarheid5. Het begin van de KOA geïnduceerd door MM is lager dan voorste cruciate ligament transect alleen of in combinatie met gedeeltelijke mediale meniscectomy6. De langdurige interacties tussen ingespoten haMSCs met de pathologische communicatie van KOA worden daarom vaak bij ratten geïnduceerd door MM7,8beoordeeld.
De therapeutische werking van haMSCs is uitgebreid gerapporteerd, relevante kennis op wel de in vivo persistentie van geïmplanteerde haMSCs via intra-articulaire (IA) injectie schaars9,10. Vandaar, diverse cellulaire labeling methoden zijn ontwikkeld, met inbegrip van de immunohistology11, luciferase12, groen fluorescent proteïne13 transfectie, ijzeroxide labeling voor magnetische resonantie beeldvorming (MRI)14 , en talrijke fluorescerende cel kleurstoffen8,15,16. Vergeleken met de arbeidsintensieve histologie analyses, in vivo niet-invasieve imaging maakt gebruik van optische apparaten te detecteren de real-time verdeling en dynamiek van cellen die zijn gelabeld met de fluorescerende signalen10,17. Voor functionele levende cel imaging is cytocompatible TL labeling een geavanceerd radioactieve-gratis tracking techniek te onthullen van cellulaire activiteiten na stamcel transplantatie18. Multicolor fluorescente lipofiele kleurstoffen bezitten bovendien voordelen ten opzichte van amino-reactieve hydrofiele kleurstoffen of fluorescerende eiwitten, met inbegrip van hun verbeterde cel permeabiliteit en verbeterde fluorescentie quantum opbrengsten19.
Dus de hier opgenomen protocollen gebruiken een rode laser te prikkelen van cellen die zijn gemarkeerd met de lipofiele carbocyanines deed (DilC,18lid 5), die is een far-red fluorescerende Dil (dialkylcarbocyanines) analoge20. De rood-verschoven excitatie en emissie spectra van vermijdt autofluorescent interferentie en diep-weefsel imaging gedurende een lange periode van tijd in levende dieren8staat. Deze methode van tracking cellen in vivo aangeduid met deed is geldig voor de controle van de getransplanteerde cellen van de stam, zoals haMSCs, in diermodellen, die is essentieel om te begrijpen en te verbeteren huidige regeneratieve therapie van de stamcel.

in vivo tracking of human adipose-derived mesenchymal stem cells in a rat knee osteoarthritis model with fluorescent lipophilic membrane dye

Ter ondersteuning van de klinische toepassing van menselijke adipeus afkomstige mesenchymale stamcellen (haMSC) therapie voor knie artrose (KOA), onderzochten we de werkzaamheid van cel persistentie en ook van haMSCs in diermodellen. We toonden een methode om de celmembraan van haMSCs met lipofiele fluorescente kleurstof van een label. Intra-articulaire injectie van de gelabelde cellen bij ratten met chirurgisch geïnduceerde KOA werd vervolgens dynamisch gecontroleerd door een in vivo imaging systeem. We gebruikt de lipofiele carbocyanines deed (DilC18 (5)), een far-red analoge tuners fluorescerende Dil (dialkylcarbocyanines) en die een rode laser gebruikt Voorkom excitatie van de natuurlijke groene autofluorescence uit de omliggende weefsels. Voorts de rood-verschoven emissie spectra van toegestaan diep-weefsel beeldvorming in levende dieren en de labeling procedure veroorzaakt geen cytotoxische effecten of functionele schade aan haMSCs. Deze aanpak is aangetoond dat een efficiënte tracking methode voor haMSCs in een rat KOA model. De toepassing van deze methode kan ook worden gebruikt om te bepalen van de optimale administratie route en de dosering van MSCs uit andere bronnen in de pre-klinische studies.

Om te induceren KOA model, werd MM uitgevoerd in het gewricht van de rechterknie van SD ratten (Figuur 3).
