Nauwkeurige meting van mitophagy is technisch veeleisend. Hier hebben wij diverse robuuste technieken waarmee beide kwalitatieve detectie van mitophagy en kwantificering van mitophagy niveaus in de meest voorkomende laboratorium experimentele modellen gepresenteerd.

Mitophagy is essentieel voor mitochondriale onderhoud. Mitochondriën kruisen meerdere cel signalering van trajecten en zijn universele sub cellulaire organellen die verantwoordelijk zijn voor cellulaire energieproductie, celstofwisseling en calcium homeostase1,2,3, 4. mitochondria voortdurend ervaring uitdagingen van endogene en exogene bronnen, zoals reactieve zuurstof soorten (ROS) en toxische stoffen in het mitochondriale, respectievelijk, die leiden tot de generatie van "leeftijd" en disfunctionele mitochondriën. Accumulatie van beschadigde mitochondriën verlaagt de efficiëntie van de ATP productie terwijl het verhogen van de hoeveelheid schadelijke ROS, en is gekoppeld aan de leeftijd gerelateerde ziekten zoals metabole ziekten, AD, en PD1,5,6 . Om te voorkomen dat mitochondriën geïnduceerde cellulaire dysfunctie, cellen moeten specifiek herkennen beschadigd mitochondriën en efficiënt verwijderen hen via een cellulaire proces genoemd mitochondriale autophagy (mitophagy). Dit toonde het belang aan van mitophagy in gezondheid en ziekte illustreert de behoefte aan nauwkeurige en efficiënte methoden voor het detecteren van mitophagy zowel in vitro als in vivo.
Mitophagy is een proces van meerdere stappen waarbij veel eiwitten en eiwit complexen5,7,8. Kortom, wordt een beschadigde mitochondrion eerst herkend en opgeslokt door een dubbel-membraned phagophore, die afkomstig van het plasmamembraan, endoplasmatisch reticulum, Golgi-complex, nucleus of mitochondrion zelf9,10 zijn kan. De sferische phagophore kieuwopeningenvorm en uiteindelijk zeehonden de mitochondriën binnen, vormen de mitochondriale autophagosome (mitophagosome). De mitophagosome combineert vervolgens met het lysosoom voor afbraak, vorming van een autolysosome waarin de beschadigde mitochondrion aangetast en gerecycleerde7,8 is. Grote autophagic eiwitten ook betrokken bij mitophagy omvatten: Autophagy verwante 7 (ATG7) en Beclin1 (initiatie), Microtubule-Associated eiwit 1A/1B-Light ketting 3 (LC3-II) (LGG-1 in C. elegans) en p62 (onderdelen van phagophore), en Lysosomale-geassocieerde membraan glycoproteïne 2 (LAMP2)6,7. Daarnaast zijn er verschillende essentiële eiwitten uniek voor mitophagy, met inbegrip van PTEN-geïnduceerde putatief Kinase 1 (roze-1), Parkin1, nucleaire Dot eiwit 52 kDa (NDP52), optineurin, BCL2 interactie eiwit 3 zoals (NIX/BNIP3L) (DCT-1 in C. elegans), o.a.5,6,11.
Een gemeenschappelijke methode voor de opsporing van wijzigingen in niveaus van autophagy is door de verhouding tussen LC3-II/LC3-I of LC3-II/actine. Deze methode is echter niet-specifieke, zoals een toename van deze verhouding kan een verhoogde initiatie of een bijzondere waardevermindering heeft ondergaan samensmelting van mitophagosome en lysosoom12weerspiegelen. Een andere methode is het evalueren van de colocalization tussen een autophagy marker (bijvoorbeeldLC3) en een mitochondriale eiwit (bijvoorbeeldTranslocase van buitenste mitochondriale membraan 20 (TOMM20, die kan worden afgebroken door Proteasomen)). Echter, dit kan alleen veranderingen in de totale mitophagy aangeven en kan geen onderscheid maken de stappen welke blokkade optreedt. Dit kan worden verduidelijkt met behulp van lysosomale remmers (bijvoorbeeld, E64d + Pepstatin A, zogenaamde EP) parallel aan de accumulatie van mitophagosomes veroorzaken. Het verschil tussen het aantal mitophagosomes op de basislijn en het aantal mitophagosomes na behandeling met EP kan duiden op mitophagy. Deze beperkingen hebben gevraagd de ontwikkeling van nieuwe mitophagy detectietechnieken. Met het oog op de toenemende relevantie van mitophagy in een breed spectrum van ziekten bij de mens presenteren wij verschillende robuuste mitophagy detectietechnieken die nuttig voor onderzoekers zijn kunnen. Wij dekken zowel in vitro als in vivo technieken en adviseren combineren meerdere technieken om te controleren of de wijzigingen van de mitophagy.

in vitro and in vivo detection of mitophagy in human cells, c. elegans, and mice

Mitochondriën zijn de krachtpatsers van cellen en cellulaire energie in de vorm van ATP produceren. Mitochondriale dysfunctie bijdraagt aan biologische veroudering en een breed scala aan aandoeningen waaronder neurodegeneratieve ziekten zoals de ziekte van Alzheimer (AD) en de ziekte van Parkinson (PD), metabole ziekten en voortijdige veroudering syndromen. Onderhoud van mitochondriale gezondheid hangt af van mitochondriale biogenese en de efficiënte goedkeuring van disfunctionele mitochondria via mitophagy. Experimentele methoden te nauwkeurig detecteren autophagy/mitophagy, met name in diermodellen, hebben zijn uitdagend om te ontwikkelen. Recente vooruitgang op weg naar het begrip van de moleculaire mechanismen van mitophagy heeft de ontwikkeling van nieuwe mitophagy detectietechnieken ingeschakeld. Hier introduceren we verschillende veelzijdige technieken voor het controleren van mitophagy in menselijke cellen, Caenorhabditis elegans (bijvoorbeeldRosella en DCT-1 / LGG-1 stammen), en muizen (mt-Keima). Een combinatie van deze mitophagy detectietechnieken, met inbegrip van cross-soorten evaluatie, zal de nauwkeurigheid van de metingen van de mitophagy verbeteren en leiden tot een beter begrip van de rol van mitophagy bij gezondheid en ziekte.

Detectie van Mitophagy in menselijke cellen:
Met behulp van de procedure die hier gepresenteerd, waren menselijke HeLa cellen transfected met mt-Keima plasmide. Gezonde cellen aangetoond een goed georganiseerde mitochondriale netwerk (GFP, 488 nm) met enkele gevallen van mitophagy (RFP, 561 nm).
