Mitofagia es esencial para el mantenimiento mitocondrial. Las mitocondrias entrecruzan múltiples vías de señalización de la célula y son organelos subcelular universales responsables de la producción de energía celular, metabolismo celular y calcio homeostasis1,2,3, 4. las mitocondrias constantemente enfrentan desafíos de fuentes endógenas y exógenas, como las especies reactivas del oxígeno (ROS) y tóxicos mitocondriales, respectivamente, que conducen a la generación de las mitocondrias disfuncionales y "envejecidas". Acumulación de mitocondrias dañadas disminuye la eficiencia de la producción de ATP mientras que aumenta la cantidad de ROS nocivos y se ha relacionado con enfermedades relacionadas con la edad, como enfermedades metabólicas, AD y PD1,5,6 . Para evitar que las mitocondrias inducido por la disfunción celular, las células necesidad reconocer específicamente dañan las mitocondrias y eliminarlos eficientemente a través de un proceso celular denominado autofagia mitocondrial (mitofagia). Mitofagia es un proceso de varios pasos en muchas proteínas y proteínas complejos5,7,8. En Resumen, una mitocondria dañada primero reconocido y envuelto por una doble membrana phagophore, que puede originar de la membrana plasmática, retículo endoplasmático, aparato de Golgi, núcleo o mitocondria sí mismo9,10. El phagophore esférico se alarga y finalmente sella las mitocondrias en el interior, constituyendo la autophagosome mitocondrial (mitophagosome). El mitophagosome entonces se fusiona con los lisosomas para su degradación, formando un autolysosome en el que la mitocondria dañada es degradada y reciclado7,8. Principales proteínas autofagia también implicadas en la mitofagia incluyen: autofagia relacionados 7 (ATG7) y Beclin1 (iniciación), proteína de Microtubule-Associated 1A/1B-luz cadena 3 LC3-II (IgG-1 C. elegans) y p62 (componentes de phagophore), y glicoproteína de membrana lisosomal asociado 2 (LAMP2)6,7. Además, hay varias proteínas esenciales exclusivo de mitofagia, incluyendo PTEN-inducida supuesta Kinase 1 (color de rosa-1), proteína Nuclear del punto 52 kDa (NDP52), Parkin1, optineurin, BCL2 interacción proteína 3 como (NIX/BNIP3L) (DCT-1 en C. elegans), entre otros5,6,11.
Un método común para detectar cambios en los niveles de la autofagia es la relación de LC3-II/LC3-I o LC3-II/actina. Sin embargo, este método es inespecífico, como un aumento en esta proporción puede reflejar una iniciación mayor o una alteración fusión de mitophagosome a lisosoma12. Otro método es evaluar la colocalización entre un marcador de la autofagia (p. ej., LC3) y una proteína mitocondrial (p. ej., Translocasa de exterior mitocondrial membrana 20 (TOMM20, que podrían ser degradados por proteasomas)). Sin embargo, esto sólo puede indicar cambios en los niveles de la mitofagia total y no puede distinguir los pasos en que bloqueo ocurre. Esto puede aclararse utilizando lysosomal inhibidores (por ejemplo, A E64d + Pepstatin, denominada EP) en paralelo para causar la acumulación de mitophagosomes. La diferencia entre el número de mitophagosomes al inicio del estudio y el número de mitophagosomes después del tratamiento con EP puede indicar mitofagia. Las mitocondrias son las centrales de las células y producen energía celular en forma de ATP. La disfunción mitocondrial contribuye al envejecimiento biológico y una amplia variedad de enfermedades incluyendo enfermedades metabólicas, síndromes de envejecimiento prematuro y las enfermedades neurodegenerativas como la enfermedad de Alzheimer (EA) y enfermedad de Parkinson (EP). Mantenimiento de la salud mitocondrial depende de la Biogénesis mitocondrial y la liquidación eficiente de las mitocondrias disfuncionales a través de mitofagia. Métodos experimentales para detectar con precisión la autofagia/mitofagia, especialmente en modelos animales, han sido un desafío para desarrollar. Recientes avances hacia la comprensión de los mecanismos moleculares de la mitofagia ha permitido el desarrollo de técnicas de detección de mitofagia novela. Aquí, presentamos varias técnicas versátiles para monitorear la mitofagia en células humanas, Caenorhabditis elegans (p. ej., Rosella y DCT-1 / IgG-1) y ratones (mt-Keima). Una combinación de estas técnicas de detección de mitofagia, incluyendo la evaluación de la especie, mejorará la precisión de las mediciones de la mitofagia y conducir a una mejor comprensión del papel de la mitofagia en salud y enfermedad.
