SH-SY5Y cellen zijn een menselijke neuroblastoma afkomstige cellijn en worden veel gebruikt als een model van de juiste cel voor in vitro onderzoek naar neurotoxiciteit of neuroprotectie12. De SH-SY5Y-cellijn was gevoelig voor glucose ontbering voorwaarden. De volwassen hersenen gebruikt bijna uitsluitend glucose als substraat voor energiemetabolisme onder normale fysiologische omstandigheden. Het menselijk brein vormt slechts 2% van het lichaamsgewicht, maar verbruikt ongeveer 25% van de totale glucose binnen het lichaam1. Het is goed gedocumenteerd dat glucose metabolisme dysfunctie is één van de belangrijkste pathologische veranderingen tijdens ischemische beroerte en diverse neurodegeneratieve ziekten, met inbegrip van de ziekte van Alzheimer (AD), de ziekte van Huntington (HD) en de ziekte van Parkinson (PD) 2,3. Het gebrek aan glucose en verminderde glucose-opname of oxidatieve fosforylatie kan rechtstreeks van invloed zijn ATP productie en verdere veroorzaken neurale celdood, die kan het risico van neuronale dysfunctie, suggereren dat handhaving levensvatbaarheid van de cellen verhogen of vertragen cel letsel na glucose ontbering misschien wel een redelijke benadering voor de behandeling van deze ziekten. Het onderzoek van neuroprotectieve effecten via glucose modulatie, gericht op ontstekingsremmers, ion kanaal modulatoren, vrije radicalen aaseters, neurotrophic factoren, etc. is van belang. Vertaling van deze neuroprotectieve benaderingen van Bank naar de klinische praktijk is echter geen succesvolle4.
Luchtdruk plasma jets (APPJs) zijn een nieuw soort atmosferische lage temperatuur gas geen kwijting technologie die heeft trok de aandacht van vele onderzoekers uit meerdere disciplines in de afgelopen jaren. APPJs zijn gebruikt voor decennia in verschillende biomedische toepassingen zoals de behandeling van de cel van de kanker, bacteriële inactivatie, bloedstolling, wondgenezing, oraal geneesmiddel, etc.5,6, als gevolg van de uitstoot tot meerdere typen van reactieve stikstof soorten (RNS) en reactieve zuurstof soorten (ROS) (Figuur 1)7. Toepassingen van de bio-geneeskunde vorige plasma is voornamelijk gericht op de oxidatieve en/of nitrative stress op bacteriën, cellen en weefsels8. De APPJ kan ook wel een "tweesnijdend zwaard" aangezien RNS en ROS belangrijke intracellulaire signalering moleculen aan vele fysiologische en pathofysiologische processen9gerelateerde zijn. Stikstofoxide (NO) beheert een breed scala aan biologische processen en speelt een dubbele rol in het menselijk lichaam, met name in het centrale zenuwstelsel (CNS). Lage niveaus van nr gebleken hun neuroprotectieve activiteiten zowel in vitro en in vivo via meerdere signaal trajecten10. Onze vorige studie eerst gemeld dat helium APPJ-geïnduceerde geen productie was betrokken bij het neuroprotectieve effect van APPJ tegen oxidatieve stress-geïnduceerde verwondingen11. De gevolgen van APPJs voor andere verwondingen zijn echter niet gemeld. Daarom is het doel van de huidige studie om een in vitro behandeling protocol betreffende de toepassingen van de neuroprotectieve van helium APPJ op glucose ontbering-veroorzaakte schade in SH-SY5Y cellen. Anders dan eerdere studies, onze protocol gebruikt lage dosis plasma behandeling voor neuroprotectieve toepassingen zonder de gevolgen van excessieve plasma-geïnduceerde verwondingen, die aangeeft dat de APPJ behandeling mogelijk zou kunnen worden gebruikt als een roman "geen donor drug "voor toekomstig onderzoek en zelfs voor klinische vertaling. Dit protocol werd ook voorgesteld om te worden gebruikt als een cytoprotective-applicatie voor andere celtypes met verschillende afboekingen, maar de APPJ behandeling voorwaarden moeten opnieuw aangepast en de dosis van de behandeling moet relatief laag.

De atmosferische druk plasma jet (APPJ) heeft trok de aandacht van vele onderzoekers uit meerdere disciplines in de afgelopen jaren, omdat de uitstoot meerdere soorten reactieve stikstof soorten (RNS) en reactieve zuurstof soorten (ROS omvatten). Onze vorige studie blijkt het effect van de cytoprotective van de APPJ tegen oxidatieve stress-geïnduceerde verwondingen. Het doel van de huidige studie is bedoeld als een gedetailleerde in vitro behandeling protocol betreffende de toepassingen van de neuroprotectieve van helium APPJs op glucose ontbering-veroorzaakte schade in SH-SY5Y cellen. De SH-SY5Y menselijke neuroblastoma afkomstige cellijn werd gehandhaafd in RPMI 1640 medium aangevuld met 15% foetaal kalfsserum. Het kweekmedium werd vervolgens gewijzigd in RPMI 1640 zonder glucose vóór de APPJ behandeling. Na een incubatie van 1 uur in een cel incubator, was de levensvatbaarheid van cellen bepaald met behulp van cel tellen Kit 8. De resultaten toonden aan dat, in vergelijking met de glucose ontbering groep, cellen behandeld met APPJ tentoongesteld aanzienlijk verhoogde levensvatbaarheid op een dosis-afhankelijke manier, met 8 s/putje waargenomen als een optimale dosis. Ondertussen, helium stroom had geen effect op de schending van de ontbering-geïnduceerde cel glucose. Onze resultaten aangegeven dat de APPJ als een behandelingsmethode van de ziekten in het centrale zenuwstelsel aan deprivatie glucose gerelateerde potentieel gebruikt kan worden. Dit protocol kan ook worden gebruikt als een cytoprotective-applicatie voor andere cellen met verschillende afboekingen, maar de celcultuur en APPJ behandeling voorwaarden moeten worden aangepast, en de dosis behandeling moet relatief laag.

Gegevens worden uitgedrukt als de gemiddelde ± SD van ten minste drie onafhankelijke experimenten. Groepsresultaten werden geanalyseerd voor variantie met ANOVA.
