SH-SY5Y celler er en human neuroblastom-avledet celle linje og er mye brukt som en aktuelle cellen modell for i vitro studier på nevrotoksisitet eller neuroprotection12. SH-SY5Y celle linjen var følsom glukose deprivasjon forhold. Voksen hjernen bruker nesten utelukkende glukose som et substrat for energi metabolism under normale fysiologiske forhold. Den menneskelige hjernen utgjør bare 2% av kroppsvekten men bruker ca 25% av den totale glukose i kroppen1. Det er godt dokumentert at glukose metabolisme dysfunksjon er en av de store patologiske forandringer under iskemiske hjerneslag og forskjellige nevrodegenerative sykdommer, inkludert Alzheimers sykdom (AD), Huntingtons sykdom (HD) og Parkinsons sykdom (PD) 2,3. Mangel på glukose og svekket glukose opptak eller oxidative fosforylering kan direkte påvirke ATP produksjonen og ytterligere indusere nevrale celledød, som kan øke risikoen for neuronal dysfunksjon, antyder at opprettholde celle levedyktighet eller forsinke celle skade etter glukose deprivasjon kan være en fornuftig tilnærming for behandling av disse sykdommene. Etterforskningen av neuroprotective effekter via glukose modulasjon, fokuserer på anti-inflammatoriske midler, ion kanal modulatorer, frie radikaler renovasjonsarbeider, nevrotropisk faktorer, etc. har vært av interesse. Oversettelse av disse neuroprotective tilnærminger fra benk til klinisk praksis har imidlertid ikke vært vellykket4.
Lufttrykk plasma jetfly (APPJs) er en ny type atmosfæriske lav temperatur gass utslipp teknologi som har tiltrukket seg oppmerksomheten til mange forskere fra flere fagfelt de siste årene. APPJs har blitt brukt i flere tiår i biomedisinsk programmer som cellen kreftbehandling, bakteriell inaktivering, blodpropp, sårheling, oral medisin, etc.5,6, på grunn av sine utslipp av flere typer reaktivt nitrogen arter (RNS) og reaktive oksygen arter (ROS) (figur 1)7. Tidligere plasma bio-medisin programmer hovedsakelig fokusert på oksidativt og/eller nitrative stress på bakterier, celler og vev8. Men kan APPJ også være en "tveegget sverd" siden RNS og ROS er viktig intracellulær signalnettverk molekylene knyttet til mange fysiologiske og patofysiologiske9. Lystgass (nei) styrer et bredt spekter av biologiske prosesser og spiller en dobbel rolle i den menneskelige kroppen, spesielt i sentralnervesystemet (CNS). Lave nivåer av ingen har vist sine neuroprotective aktiviteter både i vitro og i vivo via flere signal veier10. Vår forrige studie først rapportert at helium APPJ-indusert ingen produksjon var involvert i neuroprotective effekten av APPJ mot oksidativt stress-indusert skader11. Men er effekten av APPJs på andre skader ikke rapportert. Derfor er målet med denne studien å gi en i vitro behandling protokollen om neuroprotective programmene av helium APPJ på glukose deprivasjon-indusert skade i SH-SY5Y celler. Forskjellig fra tidligere studier, våre protokollen brukes lavdose plasma behandling for neuroprotective programmer uten konsekvensene av overdreven plasma-indusert skader, som angir at APPJ behandling potensielt kan brukes som en roman "ingen donor narkotika "for fremtidig forskning og selv for klinisk oversettelse. Lufttrykk plasma jet (APPJ) har tiltrukket seg oppmerksomheten til mange forskere fra flere fagfelt de siste årene på grunn av sine utslipp inneholder flere typer reaktivt nitrogen arter (RNS) og reaktive oksygen arter (ROS). Vår forrige studie har vist cytoprotective effekten av APPJ mot oksidativt stress-indusert skader. Målet med denne studien er å gi en detaljert i vitro behandling protokoll om neuroprotective programmene av helium APPJs på glukose deprivasjon-indusert skade i SH-SY5Y celler. SH-SY5Y human neuroblastom-avledet celle linjen ble opprettholdt i RPMI 1640 medium med 15% fosterets kalv serum. Kultur mediet ble deretter endret til RPMI 1640 uten glukose før APPJ behandling. Etter en 1t inkubasjon i en celle inkubator identifiserte celle levedyktighet bruker celle teller Kit 8. Resultatene viste at, sammenlignet med gruppen glukose deprivasjon, cellene behandles med APPJ utstilt betydelig økt celle levedyktighet i en doseavhengig måte, med 8 s/vel observert som en optimal dose. I mellomtiden hadde helium flyt ingen effekt på glukose deprivasjon-indusert celle verdifall. Resultatene indikerte at APPJ potensielt kan brukes som en behandlingsmetode for sykdommer i sentralnervesystemet knyttet til glukose deprivasjon. Denne protokollen kan også brukes som et cytoprotective program for celler med forskjellige impairments, men i cellekultur og APPJ behandling betingelsene skal justeres, og behandling dose må være relativt lav.
