SH-SY5Y celler är en human neuroblastom-derived cellinje och används allmänt som en cell lämplig modell för in vitro- studier på neurotoxicitet eller neuroprotektion12. SH-SY5Y cell fodra var känslig för glukos deprivation förhållanden. Cellernas viabilitet minskade till nästan 50% efter 1 h glukos deprivation, vilket är den optimala cell livskraft för studier av farmakodynamik. Dessutom CCK-8 reagens har ingen cytotoxicitet för celler och celler inkuberades med CCK-8 reagen...

Den vuxna hjärnan använder nästan uteslutande glukos som substrat för energiomsättning under normala fysiologiska förhållanden. Den mänskliga hjärnan utgör endast 2% av kroppsvikt men förbrukar cirka 25% av den totala glukosen i kroppen1. Det är väl dokumenterat att glukos metabolism dysfunktion är en av de stora patologiska förändringarna under ischemisk stroke och olika neurodegenerativa sjukdomar, däribland Alzheimers sjukdom (AD), Huntingtons sjukdom (HD) och Parkinsons sjukdom (PD) 2,3. Avsaknaden av glukos och nedsatt glukosupptag eller oxidativ fosforylering kan direkt påverka ATP produktionen och ytterligare framkalla neurala celldöd, vilket kan öka risken för neuronal dysfunktion, vilket tyder på att underhålla cellernas viabilitet eller att senarelägga cell skada efter glukos deprivation kan vara en rimlig strategi för behandling av dessa sjukdomar. Utredningen av neuroprotektiva effekter via glukos modulering, med fokus på antiinflammatoriska medel, ion kanal modulatorer, fri radikal renhållare, neurotrofiska faktorer, etc. har varit av intresse. Översättning av dessa neuroprotektiva strategier från bänk klinisk praxis har dock inte varit framgångsrik4.
Atmosfäriskt tryck plasma jets (APPJs) är en ny typ av atmosfäriska låg temperatur gas utsläpp teknik som på senare år har uppmärksammats av många forskare från flera discipliner. APPJs har använts i årtionden i olika biomedicinska applikationer såsom cell cancerbehandling, bakteriell inaktivering, koagulation, sårläkning, oral medicin, etc.5,6, på grund av dess utsläpp av flera typer reaktivt kväve arter (RNS) och reaktiva syreradikaler (ROS) (figur 1)7. Tidigare plasma biomedicin ansökningar inriktades främst på oxidativ och/eller nitrative stress på bakterier, celler och vävnader8. Men kan APPJ också vara ett ”tveeggat svärd” eftersom RNS och ROS är viktigt intracellulära signalmolekyler som besläktade med många fysiologiska och patofysiologiska processer9. Lustgas (NO) styr en rad biologiska processer och spelar en dubbel roll i den mänskliga kroppen, särskilt i centrala nervsystemet (CNS). Låga nivåer av nr har visat sin nervskyddande verksamhet både in vitro- och in-vivo via flera signal vägar10. Vår tidigare studie först rapporterades att helium APPJ-inducerad ingen produktion var inblandad i den neuroprotektiva effekten av APPJ mot oxidativ stress-inducerade skador11. Effekterna av APPJs på andra skador har dock inte rapporterats. Syftet med föreliggande studie är därför att ge ett in vitro- behandlingsprotokoll angående nervskyddande tillämpningar av helium APPJ på glukos deprivation-inducerad skada i SH-SY5Y celler. Skiljer sig från tidigare studier, våra protokoll används låg dos plasmabehandling för neuroprotektiva program utan konsekvenserna av överdriven plasma-inducerade skador, vilket indikerar att APPJ behandling potentiellt skulle kunna användas som en roman ”ingen givare drog ”för framtida forskning och även för kliniska översättning. Detta protokoll föreslogs också för att användas som en cytoprotektiva ansökan för andra celltyper med olika funktionsnedsättningar, men de APPJ behandling villkor bör justeras och dosen måste vara relativt låg.

Atmosfäriskt tryck plasma jet (APPJ) har uppmärksammats av många forskare från flera discipliner under de senaste åren eftersom dess utsläpp omfattar flera typer av reaktiva kväve arter (RNS) och reaktiva syreradikaler (ROS). Vår tidigare studie visat cytoprotektiva effekten av APPJ mot oxidativ stress-inducerade skador. Syftet med föreliggande studie är att ge en detaljerad i vitro behandlingsprotokoll angående nervskyddande tillämpningar av helium APPJs på glukos deprivation-inducerad skada i SH-SY5Y celler. SH-SY5Y mänsklig neuroblastom-derived cell linjen bibehölls i RPMI 1640 medium kompletteras med 15% fetalt kalvserum. Odlingssubstratet bytte sedan till RPMI 1640 utan glukos före APPJ behandling. Efter en 1 h inkubation i en cell inkubator bestämdes cellernas viabilitet med Cell räknar Kit 8. Resultaten visade att, jämfört med gruppen glukos deprivation, celler behandlas med APPJ uppvisade signifikant ökad cellviabilitet på ett dosberoende sätt, med 8 s per brunn observerats som en optimal dos. Under tiden hade helium flöde ingen effekt på glukos deprivation-inducerad cell nedskrivning. Våra resultat visade att APPJ potentiellt skulle kunna användas som en behandlingsmetod för sjukdomar i centrala nervsystemet relaterade till glukos deprivation. Detta protokoll kan också användas som ett cytoprotektiva program för andra celler med olika funktionsnedsättningar, men de cellkultur och APPJ behandling villkor bör justeras dosen måste vara relativt låg.
