Muchas proteínas están implicadas en el enlace de G4 y regulación, incluyendo proteínas telomere (telomerasa, TRF2, POT1, RPA, TEBPs)1,2, factores de transcripción (nucleolin PARP1)3, RNA, proteínas (hnRNP A1, de procesamiento hnRNP A2)4, helicasas (BLM FANCJ RHAU, WRN, Dna2, Pif1)5y replicación del ADN relacionadas con proteínas (Rif1, REV1, PrimPolymerase)6. Unión a proteínas puede estabilizar o desestabilizar las estructuras G4; así regular las funciones biológicas posteriores. La estabilidad del G4 se midió por termo fusión usando radiación ultravioleta (UV) o métodos de dicroísmo circular (CD)7. El rápido desarrollo de tecnologías de manipulación de una sola molécula ha permitido estudios de plegar y desplegar de una biomolécula, como ADN o una proteína, a nivel de una sola molécula con resolución nanométrica en tiempo real8. Microscopía de fuerza atómica (AFM), pinzas ópticas y pinzas magnéticas son los más utilizados métodos de manipulación de una sola molécula. En comparación con AFM y pinza óptica9, pinzas magnéticas permiten mediciones estables de plegar-desplegar dinámicas de una sola molécula durante días utilizando una técnica anti deriva10,11.
Aquí, una plataforma de manipulación de una sola molécula con pinzas magnéticas para estudiar la regulación de la estabilidad de la G4 de las proteínas es divulgado12,13. Este trabajo describe los métodos básicos, incluyendo preparación de muestra y el flujo de canal, la instalación de pinzas magnéticas y la calibración de fuerza. Permiten el control de la fuerza y los protocolos anti deriva como se describe en el paso 3 para largo tiempo las mediciones con varios controles de la fuerza, como fuerza constante (fuerza de sujeción) y constante carga tarifa (fuerza-rampa) y medición de salto de fuerza. Estructura secundaria de ácidos nucleicos no canónicos que g-quadruplexes (G4) intervienen en diversos procesos celulares, tales como la replicación del ADN, transcripción, procesamiento de RNA y alargamiento de los telómeros. Durante estos procesos, varias proteínas se unen y resolución estructuras G4 para realizar su función. Como la función de G4 a menudo depende de la estabilidad de su estructura doblada, es importante investigar cómo G4 proteínas regulan la estabilidad del G4. Este trabajo presenta un método para manipular las moléculas individuales de G4 con pinzas magnéticas, que permite estudios de la regulación de proteínas G4 en una sola molécula G4 en tiempo real.
