De ORA kunt de verfijnde functionele analyse van de twee wezenlijke basiscomponenten van de intestinale stamcel niche, namelijk Lgr5+ en Paneth cellen. Het intestinaal epitheel is is het snelst zichzelf vernieuwende weefsel in het lichaam van zoogdieren en, als zodanig, het object van een overvloed aan studies gericht op de identificatie en functionele karakterisering van de volwassen stamcellen, wonende aan de onderkant van de crypte van Lieberkühn, gereserveerde door expressie van het Lgr5 -gen en afhankelijk van de canonieke Wnt signalen1. Met name zijn de cellen van de stam van de Lgr5+ geflankeerd en ondersteund door gespecialiseerde niche cellen, d.w.z. de Paneth cellen, die ook afhankelijk zijn van de Wnt signalering voor hun rijping2. Samen, deze twee celtypes ten grondslag liggen aan de zelf-vernieuwing van de epitheliale darmwand en het dagelijkse homeostatische evenwicht bewaren: Lgr5+ stamcellen snel verdelen en aanleiding geven tot voorlopercellen en meer gespecialiseerde intestinale epithelial cellen; Paneth cellen essentiële niche factoren (bijvoorbeeld, Dll1, Wnt3, EGF) geven Lgr5+ 2van de cellen van de stam. De capaciteit van intestinale crypten wanneer vergulde ex vivo vormen van georganiseerde en zichzelf vernieuwend structuren organoids, of "mini-guts", genoemd als een experimenteel hulpmiddel om inzicht in processen zoals zelf-vernieuwing is benut en differentiatie in normale en pathologische omstandigheden waaronder kanker4. Organoid culturen zijn verschillende weefsel, met inbegrip van de darm, de alvleesklier, de lever, en de nier, zowel muis als menselijke specimens4vastgesteld. De extractiemethode en de groeifactoren werkzaam te ontwikkelen deze organoid culturen zijn weefsel-specifieke en ontworpen om te rijden meerdere lineage differentiatie en bootsen zo dicht mogelijk de oorspronkelijke cel van de stam-niche in vivo. Organoids wellicht potentiële toepassingen met inbegrip van de behandeling van genetische ziekten, de beoordeling van de therapeutische werking in kanker, de analyse van de drug toxiciteit of de studie van de organogenese in vitro5.
De belangrijkste beperking van organoid culturen als van weefselmonsters tot stand gebracht is over het algemeen een gebrek cel-specificiteit. Bijvoorbeeld, intestinale organoids vastgesteld van hele intestinale crypten niet toestaan de analyse van individuele cellulaire componenten omvatte binnen de bron van het weefsel (bvhele crypten bevatten Lgr5+, Paneth, en voorlopercellen).
Hier beschrijven we een nieuwe methode, hierna aangeduid als ORA, die de voordelen van intestinale organoid culturen met de functionele analyse van de meest fundamentele onderdelen, namelijk stam (Lgr5+) en niche (Paneth) cellen combineert. Dit wordt bereikt door het unieke vermogen van Paneth en Lgr5+ cellen om fysiek te koppelen aan elkaar wanneer mede geïncubeerd en aanleiding tot organoids2,6,10 geven. Wij maakte gebruik van deze functie en vooraf behandeld de twee celtypes individueel alvorens deze te reconstrueren organoids. Daarbij elk onderdeel van de cel kan worden blootgesteld aan bepaalde drug, groeifactor, biochemische remmer, genetische modificatie, of chemische behandeling voorafgaand aan de reconstructie en de vorming van de organoid. Met behulp van de ORA bepaling staat dan ook toe de bepaling van de vraag of een specifieke medicamenteuze behandeling of genetische modificatie een bepaald effect op de cellen van de stam of de wederpartij van hun niche heeft.

Het intestinaal epitheel wordt gekenmerkt door een zeer snelle Verwerkingsfrequentie. Bij zoogdieren, wordt de gehele epitheliale bekleding vernieuwd binnen 4-5 dagen. Volwassen intestinale stamcellen bevinden zich aan de onderkant van de crypten van Lieberkühn, door de expressie van het Lgr5 -gen zijn bestemd en homeostase via hun karakteristieke proliferatieve hoog1behouden. In de dunne darm, Lgr5+ stamcellen zijn vermengd met gespecialiseerde secretoire cellen genaamd Paneth cellen. Paneth cellen afscheiden van antibacteriële stoffen (d.w.z., lysozym en cryptdins/defensinen) en het uitoefenen van een controlerende rol op de darmflora. Meer onlangs, is een nieuwe functie voor Paneth cellen, namelijk hun capaciteit niche steun bieden aan de cellen van de stam van Lgr5+ door middel van verschillende belangrijke liganden zoals Wnt3, EGF en Dll12ontdekt.
Als ex vivo geïsoleerd en in aanwezigheid van bepaalde groeifactoren en componenten van de extracellulaire matrix gekweekt, hele intestinale crypten aanleiding geven tot langlevende en zelf vernieuwen 3D structuren genoemd organoids die sterk lijken op de crypt-villosités epitheliale architectuur van de volwassen dunne darm3. Organoid culturen, als tot stand gebracht van hele crypten, toestaan dat de studie van zelf-vernieuwing en differentiatie van de intestinale stamcel niche, maar zonder een aanpak van de bijdrage van de afzonderlijke onderdelen, namelijk de Lgr5+ en Paneth cellen.
Hier beschrijven we een nieuwe benadering voor de organoid-bepaling die van de mogelijkheid van Paneth profiteert en Lgr5+ cellen vormen van organoids te koppelen wanneer mede gekweekt. Deze aanpak, aangeduid als de "organoid reconstitutie assay" (ORA), kan de genetische en biochemische wijziging van Paneth of Lgr5+ stamcellen, gevolgd door reconstitutie in organoids. Als zodanig, maakt het de functionele analyse van de twee belangrijkste componenten van de intestinale stamcel niche.

De bepaling van de reconstitutie organoid kunt de aparte functionele analyse van de essentiële onderdelen van de niche en cel van de stam van het intestinaal epitheel, hier gedemonstreerd door Klein Mengend RNA (siRNA) van de Apc -gen.
