Hoewel optogenetic tools veel gebruikt worden om te studeren ziekten, en fysiologie bij knaagdieren, heeft de technische uitdaging van verhelderend grote hersenen volumes beperkt het gebruik van optogenetics in niet-menselijke primaten. Baanbrekende studies in apen grote lichte macht dichtheden (~ 100 mW/mm2 tot 20 W/mm2) gebruikt voor het verlichten van kleine volumes, misschien &#60; 1 mm3en gerapporteerde bescheiden gedrags effecten met excitatory opsins in de cortex4...

Optogenetic instrumenten, die voor milliseconde-nauwkeurige, licht gestuurde neuronale controle zorgen worden veel gebruikt om te studeren functionele fysiologie en gedrag in knaagdieren en ongewervelde dieren. Technische uitdagingen hebben echter slechts beperkt het gebruik van optogenetics in de hersenen van de niet-menselijke primaten, die een volume van ~ 100 x groter dan de knaagdier hersenen 1 heeft.
Om te vergemakkelijken optogenetics studies in niet-menselijke primaten, een hulplicht is ontworpen om twee concurrerende doelstellingen adres: groot volume verlichting en minimale penetratie schade. Eerdere pogingen om een van deze problemen te komen op de duur van de andere. Bundels van vezels verlichten grotere volumes, maar met grotere diameter, en daarmee schade2,3. Taps toelopende glasvezels beschadiging, penetratie, maar ternauwernood focus licht aan oplichtende oppervlak &#60; 100 µm2 - 4,5. Externe hersenen verlichting door een raam in de dura omzeilt de uitdaging van de penetratie schade en kan voor grote volume verlichting, maar het kan alleen worden gebruikt voor enkele oppervlakkige hersenen gebieden6.
Als u wilt maken een groot volume, kleine diameter illuminator (Figuur 1a), het puntje van een plastic optische vezel is warmte tapered en de kern en de bekleding zijn geëtst (Figuur 1bc). In tegenstelling tot andere taps toelopende vezels die licht naar een smal punt concentreren, kunt de etsen licht gelijkmatig ontsnappen uit de zijkanten van de stortplaats, dus licht in grote lijnen over een groot gebied (Figuur 1 de) te verdelen. Omdat de penetratie schade is evenredig aan de diameter van de penetratie, dit hulplicht heeft geen meer schade van de penetratie dan een conventionele vezel, maar het heeft &#62; 100 x het oplichtende oppervlak gebied en levert licht meer in het algemeen met 1/100ste de lichte macht dichtheid in een hersenen phantom (1,75% agarose) (Figuur 1e). Een Monte Carlo-model (Figuur 1f) illustreert het verschil in licht verspreiden tussen een conventionele vezel en het grote volume hulplicht wanneer zij gelijke lichte macht dichtheden hebben als hun lichtdoorlatende oppervlakken. Elke illuminator is individueel gekalibreerd met behulp van een bolfotometer (Figuur 2a, b) om ervoor te zorgen zelfs lichtverdeling langs de tip (Figuur 2 c).
Dit groot volume illuminator is gevalideerd met optogenetic manipulatie van zowel gedrag en neuronale afvuren in niet-menselijke primaten. De lengte van de tip vezel kan worden aangepast naar elk willekeurig deel van de hersenen en voor elk dier individuele receptieve veld kaart. Het hulplicht kan gekoppeld worden aan een doordringende elektrode voor neuronale opnames die zich uitstrekken over de lengte van de verlichting. Verder, omdat de vezel elke kleur van zichtbaar licht kunnen, kunnen worden gekoppeld met een van de beschikbare optogenetic moleculen beschikbaar.

Dit protocol beschrijft een groot volume illuminator, die werd ontwikkeld voor optogenetic manipulaties in de hersenen niet-menselijke primaten. De illuminator is een gemodificeerde kunststof optische vezel met geëtste tip, zodanig dat het oplichtende oppervlak &#62; 100 x die van een conventionele vezel. Naast het beschrijven van de bouw van een groot volume hulplicht, detailleert dit protocol de kalibratie van de kwaliteitscontrole gebruikt om ervoor te zorgen zelfs lichtverdeling. Dit protocol wordt verder beschreven technieken voor het invoegen en verwijderen van het hulplicht groot volume. Zowel de oppervlakkige en de diepe structuren kunnen worden verlicht. Dit hulplicht groot volume hoeft niet te worden fysiek gekoppeld aan een elektrode, en omdat de illuminator is gemaakt van kunststof, niet het glas, het gewoon zal buigen in omstandigheden als traditionele optische vezels zou versplinteren. Omdat dit hulplicht licht boven gedragsgestoorde relevante weefsel volumes (≈ 10 mm3) zonder meer schade van de penetratie dan een conventionele optische vezel levert, vergemakkelijkt het behavioral studies met optogenetics in niet-menselijke primaten.

De verlichting van de grote hersenen volumes in niet-menselijke primaten zorgt voor behavioristisch relevante optogenetic manipulatie. Acker et al. (2016) gebruikt dit hulplicht groot volume met de rood-verschoven Halorhodopsin, Jaws, 7 te bestuderen van de tijdelijke bijdrage van het veld frontale oog (FEF) geheugen-geleide saccades in twee rhesus monkeys.
