Alors que les outils d’optogenetic sont largement utilisés pour étudier la maladie et la physiologie chez les rongeurs, le défi technique de volumes de gros cerveau éclairante a limité l’utilisation des optogenetics chez les primates non humains. Optogenetic outils, qui permettent un contrôle neuronal milliseconde-précis et axée sur la lumière sont largement utilisés pour étudier la physiologie fonctionnelle et le comportement des rongeurs et des invertébrés. Toutefois, des défis techniques ont limité l’utilisation d’optogenetics dans le cerveau de primate non humain, qui a un volume ~ 100 x plus grand que le cerveau de rongeurs 1.
Pour faciliter les études d’optogenetics chez les primates non humains, enlumineur a été conçu pour répondre à deux objectifs contradictoires : illumination de grand volume et dommages de pénétration minimale. Les tentatives précédentes pour relever l’un de ces préoccupations sont venus à la coûteuse de l’autre. Faisceaux de fibres s’allument plus grands volumes mais avec diamètre augmenté et, par conséquent, dommages2,3. Fibre de verre coniques réduire les dommages de pénétration, mais étroitement mise au point de lumière émettant des surfaces &#60; 100 µm2 4,5. Illumination de cerveau externe à travers une vitre dans la dure-mère contourne le défi des dommages de la pénétration et peut permettre pour l’éclairage de volume important, mais il seulement peut être utilisé pour quelques de zones cérébrales superficielles6.
Pour créer un illuminateur de grand volume, petit diamètre (Figure 1 a), la pointe d’un plastique optique fibre est conique de la chaleur et le noyau et le revêtement sont gravés (Figure 1 bc). Contrairement à d’autres fibres coniques qui se concentrent la lumière à un point précis, la gravure permet à la lumière de s’échapper uniformément sur les côtés de la pointe, ainsi, distribuer la lumière largement sur une grande surface (Figure 1 de). Parce que les dommages de pénétration sont proportionnel au diamètre de la pénétration, ce projecteur a aucun dommage de pénétration plus qu’une fibre conventionnelle, pourtant, il a &#62; 100 x la lumière émettant une lumière de zone et livre surface plus large avec 1/100e la puissance lumineuse densité dans un cerveau fantôme (1,75 % d’agarose) (Figure 1e). Un modèle Monte Carlo (Figure 1f) illustre la différence dans la propagation de lumière entre une fibre conventionnelle et l’illuminateur de grand volume lorsqu’ils ont des densités de puissance lumineuse égale comme leur éclairantes. Chaque projecteur est calibré individuellement à l’aide d’une sphère d’Ulbricht (Figure 2 a, b) pour assurer la distribution même la lumière le long de la pointe (Figure 2c).
Ce projecteur grand volume a été validé avec optogenetic manipulation du comportement et de décharge neuronale chez les primates non humains. La longueur de pointe de fibre peut-être être personnalisée à une zone du cerveau et au mappage de champ réceptif individuels de chaque animal. L’illuminateur peut être couplé avec une électrode pénétrante pour les enregistrements neuronales qui s’étendent sur la durée de l’éclairement. En outre, parce que la fibre peut porter n’importe quelle couleur de la lumière visible, il peut être couplé avec n’importe lequel des molécules disponibles optogenetic disponibles.

Ce protocole décrit un illuminateur de grand volume, qui a été développé pour des manipulations d’optogenetic dans le cerveau de primate non humain.
