Ferramentas de Optogenetic, que permitem controle neuronal milissegundo de precisão, luz-driven são amplamente utilizadas para estudar a fisiologia funcional e comportamento em roedores e invertebrados. No entanto, os desafios técnicos limitaram-se a utilização de optogenetics no cérebro de primatas não humanos, que tem um volume de ~ 100 x maior do que o cérebro de roedor 1.
Para facilitar estudos de optogenetics em primatas não-humanos, um iluminador foi projetado para atender a dois objetivos concorrentes: iluminação de grande volume e dano mínimo de penetração. Tentativas anteriores de endereço dentre estas preocupações vir no cara do outro. Feixes de fibras iluminar volumes maiores, mas com diâmetro maior e, assim, danos2,3. Fibras de vidro cônico reduzem danos de penetração, mas por pouco o foco de luz emitindo áreas de superfície &#60; 100 µm2 4,5. Iluminação externa do cérebro através de uma janela na dura-máter contorna o desafio dos danos de penetração e pode permitir a iluminação de grande volume, mas ele só pode ser usado para algumas áreas de cérebro superficial6.
Para criar um iluminador de grande volume, pequeno diâmetro (Figura 1a), a ponta de um plástico óptico de fibra é calor afilado e o núcleo e o revestimento são gravadas (Figura 1bc). Ao contrário de outras fibras afiladas que focam a luz para um ponto estreito, a gravura permite que a luz uniformemente escapar para fora dos lados da ponta, assim, distribuir luz amplamente sobre uma grande área (Figura 1 de). Porque o dano de penetração é proporcional ao diâmetro de penetração, este iluminador não tem nenhum dano de penetração mais do que uma fibra convencional, ainda tem &#62; 100 x a luz emitindo luz superfície de área e oferece mais amplamente com 1/100 o poder da luz densidade em um cérebro fantasma (1,75% do agarose) (Figura 1e). Um modelo de Monte Carlo (Figura 1f) ilustra a diferença na propagação de luz entre uma fibra convencional e o iluminador de grande volume, quando eles têm densidades de energia de luz igual como sua superfícies de emissores de luz. Cada iluminador é calibrado individualmente usando uma esfera de Ulbricht (Figura 2a, b) para assegurar a distribuição de luz até ao longo da ponta (Figura 2C).
Este iluminador de grande volume foi validado com manipulação de optogenetic de comportamento e de disparo neuronal em primatas não-humanos. O comprimento da ponta de fibra pode ser personalizado para qualquer área do cérebro e mapa de campo receptivo individual de cada animal. O iluminador pode ser emparelhado com um eletrodo penetrante para gravações neuronais que abrangem o período de iluminação. Além disso, porque a fibra pode carregar qualquer cor de luz visível, pode ser emparelhado com qualquer das moléculas optogenetic disponível disponíveis.
