המטרה של פרוטוקול זה הוא לבודד את וירוס הרפס סימפלקס סוג 1 (HSV-1) ה-DNA של תאים נגועים לצורך הזיהוי הקשורים ויראלי החלבונים על ידי מסה ספקטרומטר. למרות חלבונים המקיימים אינטראקציה עם הגנום הנגיפי משחק תפקידים עיקריים בקביעת התוצאה של זיהום, ניתוח מקיף של גנום ויראלי הקשורים חלבונים לא היה אפשרי בעבר. כאן נדגים שיטה המאפשרת את טיהור ישירה של HSV-1 הגנום של תאים נגועים. שכפול ה-DNA הנגיפי מסומן באופן סלקטיבי עם נוקלאוטידים ששונה מכילים קבוצה פונקציונלית של אלקין. דנ א שכותרתו אז במיוחד ובלתי הפיך מתויג באמצעות החזקה קוולנטיות של ביוטין אזיד דרך נחושת (I)-מזורז אזיד-אלקין cycloaddition או לחץ על התגובה. מתויג ביוטין DNA מטוהר על חרוזים מצופים streptavidin, חלבונים הקשורים eluted, המזוהה באמצעות ספקטרומטר מסה. שיטה זו מאפשרת מיקוד סלקטיבי ובידוד של מזלגות שכפול HSV-1 או כל הגנום של סביבות ביולוגיות מורכבות. יתר על כן, עיבוד של גישה זו יאפשר החקירה של היבטים שונים של זיהום herpesviral, כמו גם הבחינה של הגנום של וירוסים אחרים, ה-DNA.
