Toutefois, la plupart des nanostructures synthétisés par cette méthode sont principalement plafonnées avec des ligands hydrophobes tels que l’acide oléique et oleylamine, qui empêchent généralement leur bioapplication supplémentaire en raison de la limited de solubilité de ligand hydrophobe en solution aqueuse 15. par conséquent, il est nécessaire d’effectuer des techniques de modification de surface convenable pour préparer UCNs biocompatibles en applications biologiques in vitro et in vivo.

synthesis of core-shell lanthanide-doped upconversion nanocrystals for cellular applications

Lanthanide dopé upconversion nanocristaux (UCNs) ont attiré beaucoup d’attention ces dernières années, selon leurs propriétés optiques prometteuses et contrôlables, qui permettent l’absorption de lumière de proche infrarouge (NIR) et peuvent ensuite convertir en multiplexage des émissions qui s’étendent sur un large éventail de régions de l’UV à la visible pour le RNI. Cet article présente les procédures expérimentales détaillées pour la synthèse de la coprécipitation de haute température du noyau-enveloppe UCNs qui intègrent des ions lanthanides différents dans les nanocristaux pour convertir efficacement excitation de NIR perméables aux tissus profonds (808 nm) dans une forte émission de bleue à 480 nm. En contrôlant la modification de surface avec polymère biocompatible (acide polyacrylique, AAP), l’UCNs préparés comme acquiert grande solubilité dans les solutions tampons. Les nanocristaux hydrophiles sont davantage fonctionnalisés avec des ligands spécifiques (cyclooctyne de dibenzyle, DBCO) pour la localisation sur la membrane cellulaire. À la lumière NIR (808 nm) irradiation, l’émission d’upconvertis bleu peut activer efficacement la protéine canal lumière bloquées sur la membrane cellulaire et réglementer spécifiquement l’afflux du cation (par exemple, Ca2 +) dans le cytoplasme. Ce protocole prévoit une méthode possible pour la synthèse du noyau-enveloppe dopé lanthanide UCNs et ultérieure biocompatible modification de surface pour des applications cellulaires plus.

Le processus de synthèse schématique du noyau-enveloppe dopé lanthanide UCNs sont indiquées dans la Figure 1 et Figure 2.
