1. bloem en Hauw fixatie

Bloemen zijn een van de belangrijkste organen van bedektzadigen definiëren. Bloeiende planten ongeveer 90-130 miljoen jaar geleden verscheen1, en gediversifieerde zo snel dat hun snelle verschijning werd beschreven als een "verschrikkelijke verborgenheid" door Charles Darwin2. De belangen van plant onderzoekers in Bloemontwikkeling zijn divers. Wat onderzoek heeft zich gericht op het begrijpen van de evolutionaire oorsprong van de bloem als geheel, of de ontwikkeling van specifieke anatomische, structurele en functionele eigenschappen van bloemen3,4,5,6 . De hoge variatie in floral vorm en structuur, evenals de vervoermiddelen seksuele en ongeslachtelijke voortplanting beroep op hen, maken de bloem een zeer complexe structuur. Dit heeft geleid tot diverse inspanningen te karakteriseren van de anatomie en de structurele kenmerken van floral organen, met behulp van licht en elektron microscopische technieken die kunnen worden gecombineerd met genetische en moleculaire onderzoeken7. Bovendien, als de bron van vruchten en zaden, zijn bloemen van het allergrootste belang voor de voeding van mens en dier. Dus, de karakterisatie van bloem en fruit ontwikkeling heeft veel gevolgen voor toegepast onderzoek, met inbegrip van de veiligstelling van de voedselvoorziening voor een toenemende menselijke bevolking en de strategieën van de ecologische behoud onder een veranderende omgeving8 , 9 , 10.
Bloemontwikkeling Arabidopsis begint met bloem inductie en de transformatie van de vegetatieve meristeem aan een meristeem bloeiwijze (groep van bloemen). Bloem primordia zijn lateraal gestart op de flank van de bloeiwijze meristeem11. De bloemen orgel primordia vormen geleidelijk in concentrische slierten van buiten naar het midden van de bloem, en zich uiteindelijk ontwikkelen tot kelkbladen, bloemblaadjes, meeldraden en carpellen7. Deze bloemen organen vervullen verschillende voedend, beschermende en functionele (bijvoorbeeldpollinator attractie) rollen in verschillende plantensoorten, met het ondersteunen van de ontwikkeling van mannelijke en vrouwelijke gametofyten, respectievelijk12 geslachtsorganen , 13. de gametofyten, op zijn beurt, elk onderscheiden een paar van mannelijke (sperma) en vrouwelijke gameten (ei en centrale cel), die verenigen op dubbele bevruchting tot het vormen van de volgende generatie, de zygote en de primaire endosperm, een terminal weefsel, ter ondersteuning van de ontwikkeling van het embryo14,15. Vruchten en zaden ontwikkeling steunen de groei, rijping en bewaring van het embryo en, uiteindelijk, de versnippering. Uitgebreid onderzoek heeft verricht om het karakteriseren van bloem en embryo-ontwikkeling in diverse plantensoorten, vooral in de model soorten Arabidopsis7,16,17.
Vroege microscopische analyses van de bloemontwikkeling waren gebaseerd op tijdrovende monster verwerking en observatie technieken, zoals paraffine of hars inbedding en segmenteren, in combinatie met licht of elektronenmicroscopie. Deze traditionele microscopische technieken werden vaak gebruikt in combinatie met moleculaire genetische onderzoeken, zoals microscopische analyses van mutanten, de lokalisatie van RNA door in situ hybridisatie, of de immuno-detectie van eiwitten. Recente vooruitgang in breed-gebied en confocal fluorescentie microscopie, in de state-of-the-art 3-D computer-aided beeld analyses, en de continue verfijning van moleculaire methoden die kunnen worden gebruikt op minimaal verwerkte geheel-mount monsters, heeft geleid tot een behoefte aan efficiënte en minimal steekproef verwerkingstechnieken die bij voorkeur zich lenen voor kwantitatieve analyses. In de afgelopen jaren aanzienlijke vooruitgang geboekt bij de ontwikkeling van clearing technieken op geheel-mount dierlijke exemplaar. Zij maken het monster transparant door gebruik te maken van waterige ureum of suiker gebaseerde reagentia (bijvoorbeeld, schaal, SeeDB, CUBIC)18,19,20, of door het selectief verwijderen van lipiden (met het detergent SDS) na monsters te Embedding in stabiele hydrogels; de verwijdering van lipiden kan worden bereikt door passieve diffusie (b.v.gewijzigd duidelijkheid protocol21, PACT-PARS-velgen22) of actief door elektroforese (oorspronkelijke duidelijkheid protocol23 en ACT-PRESTO-24). Aangemoedigd door deze snelle vooruitgang, enkele afgeleide technieken zijn ook in opkomst voor gebruik in planten.
In deze paper methoden gericht op het model Arabidopsis, beschrijven we de procedure voor de juiste dissectie van de bloemknoppen, bloemen, en jonge siliques en het opruimen van geheel-mount monsters voor uiteenlopende kleuring en observatie procedures met klassieke of een recente SDS gebaseerde clearing methode. Voorbeelden voor zetmeel, callose en chromatine kleuring worden gegeven. Hoewel deze procedures verdere wellicht verbeteringen en aanpassingen bij gebruik in combinatie met andere soorten, we hopen dat ze zal het decor voor verder onderzoek op deze eenvoudige maar kritische methoden die het startpunt van vele onderzoeksprojecten.

Echter, plant morfologische, anatomische en ultrastructurele analyses traditioneel betrekken tijdrovende insluiten en segmenteren van de procedures voor heldere veld, scannen en elektronenmicroscopie. Recente vooruitgang in confocale fluorescentie microscopie, state-of-the-art 3-D computer-aided microscopische analyses en de continue verfijning van moleculaire technieken om te worden gebruikt op minimaal verwerkte geheel-mount monsters, heeft geleid tot een toegenomen vraag naar de ontwikkeling van efficiënte en minimal steekproef verwerkingstechnieken.
