本稿では,レプトスピラ変異ゴールデン ハムスター感染中フィットネスに変更の定量化 (Tn Seq) 手法をシーケンス トランスポゾンをについて説明します。Tn Seq は高スループット シーケンス技術の力とランダムなトランスポゾン変異を兼ね備えています。動物は収穫血液と組織の数日後、各器官 (出力プール) の突然変異体の数を定量化することによって続いてトランスポゾン変異体 (入力プール) のプールに立ち向かっています。出力のプールは、各変異体の生体内で適性を評価する入力のプールと比較されます。このアプローチにより、動物の限られた数の突然変異体の大規模なプールのスクリーニングです。マイナーな変更とレプトスピラ症の動物モデル、ラットなど貯留層ホスト モデルとの in vitro研究と同様、ハムスターなどの急性感染症モデルでこのプロトコルを実行できます。Tn Seq は、体内と体外のフィットネス欠陥を持つ突然変異体の画面に強力なツールを提供します。
