Het bovenstaande protocol beschrijft een strategie voor het verkrijgen van kwantitatieve gegevens uit afbeeldingen van hoge kwaliteit hele mount immunofluorescentie van de muis na implantatie embryo's. De studie van de organogenese heeft lang vertrouwd op waarneming van morfogenetische gebeurtenissen in het zich ontwikkelende embryo. Deze studies is vaak afhankelijk van het gebruik van fluorescente kleurstoffen of lineage tracering verslaggevers in combinatie met het labelen van gedefinieerde referentie populaties. 1 door de relatieve posities van deze labels te vergelijken, informatie kan worden opgedaan op de oorsprong, beweging of uiteindelijke bijdrage van een bevolking van belang. Transplantatie en lot toewijzing experimenten kunnen morfologische monumenten of injectie van verf in niet-sporenvormende lineages definiëren van het startpunt van de cellen van belang, die vervolgens voor bijdrage aan het ontwikkelde embryo worden onderzocht. 2 , 3 , 4 , 5 genetische afkomst-tracing experimenten gebruik hetzelfde concept met welomschreven verslaggever allelen die worden gebruikt om de label-cel populaties zonder experimentele manipulatie. Sleutel tot deze benaderingen is de mogelijkheid om te bepalen, met hoge ruimtelijke resolutie, de locaties van de experimentele en verwijzing labels. De ontwikkelingstoxiciteit gebeurtenissen die ten grondslag liggen aan hart morfogenese zijn steeds goed beschreven in de afgelopen jaren geweest. 10 één van de belangrijke ontdekkingen op het gebied van onderzoek is de beschrijving van een aantal voorlopercellen populaties die kunnen worden onderscheiden door de expressie van unieke markers. 11 deze populaties zijn de eerste en de tweede hart velden (FHF en SHF), die aanwezig is binnen de cardiale halve maan aan de voorste zijde van het embryo ten embryonale dage (E) 8,25 van muis ontwikkeling zijn. 12 deze populaties worden vaak onderzocht door een combinatie van breed-gebied microscopie, waarmee weefsel-niveau informatie, en seriële afdelen met immunofluorescentie testen, die hoge cellulaire resolutie maar alleen biedt tweedimensionale ruimtelijke informatie. 13 aldus, terwijl deze studies zeer ons begrip van de ontwikkeling van het hart geavanceerde hebben, de beschikbare methoden hebben beperkte diepgaande kwantitatieve analyse van genuitdrukking tijdens deze fasen, maken de noodzaak voor benaderingen te onderzoeken de organisatie van deze populaties op het niveau van een geheel-organisme.
De recente vooruitgang in zowel confocale microscopie en 3D beeldanalyse voorzien in een hoge resolutie en hoge gegevensdoorvoer algoritmische reconstructies van de cellen en structuren in situ met relatief gemak, dus de weg vrijmaakt voor gedetailleerde studies van complex cellulaire structuren. 14 met de toename van rekenkracht en de ontwikkeling van grote-data beheren algoritmen, beide nodig om de exponentiële toename van de grootte van de imaging gegevenssets, analyses kunnen nu volledig geautomatiseerd worden. 15 geautomatiseerde analyse van beeldvorming-gegevensverzamelingen, heeft het voordeel dat onbevooroordeelde, maar het is alleen zo betrouwbaar als de kwaliteit van de input dataset; het is dan ook absoluut noodzakelijk dat best practices worden gebruikt tijdens de verwerving en voorbehandeling van de afbeelding om de hoogste kwaliteit, onbevooroordeelde analyse. 16 kunnen protocollen volledig geautomatiseerd en gedeelde voor de reproduceerbaarheid en de algoritmen die worden gebruikt door propriëtaire software gemakkelijk beschikbaar zijn via bibliotheken worden gebruikt door wetenschappers hebben vertrouwdheid met moderne farmaceutische of open-source Ontwikkelaarshulpmiddelen. 17
Het volgende protocol legt de nodige stappen om dergelijke analyses uit te voeren op één welbepaalde model van organogenese, de vorming van de cardiale halve maan tijdens de ontwikkeling van het hart. Specifiek, dit protocol wordt beschreven hoe u (1) oogst en ontleden van cardiale halve maan fase embryo's, (2) uit te voeren gehele mount immunofluorescentietest voor verwijzing (Nkx2-5) en experimentele (Foxa2Cre:YFP18,19) markeringen, (3) bereiden en beeld van de embryo's met behulp van de confocal microscopie, en ten slotte (4) analyseren en kwantificeren van de resulterende afbeeldingen met behulp van geavanceerde driedimensionale benaderingen. Het gebruik van steeds voortschrijdende beeldvormingstechnieken heeft over het algemeen bijgedragen aan onze toegenomen kennis van embryonale ontwikkeling. Pre-implantatieontwikkeling en organogenese zijn twee gebieden van onderzoek die sterk van deze voorschotten, als gevolg van de hoge kwaliteit van de gegevens die kan worden verkregen profiteerden hebben vanuit imaging pre-implantatie embryo's of ex vivo organen. Pre-implantatie embryo's hebben opgeleverd gegevens met vooral hoge ruimtelijke resolutie, zijn latere stadia geweest minder vatbaar voor driedimensionale wederopbouw. Kwalitatief hoogwaardige 3D- of volumetrische gegevens voor bekende embryonale structuren in combinatie met lot toewijzing of genetische afkomst tracering te verkrijgen zal zorgen voor een meer uitgebreide analyse van de morfogenetische evenementen die plaatsvinden tijdens de embryogenese.
Dit protocol beschrijft een geheel-mount immunofluorescentie aanpak waarmee de labeling, visualisatie en kwantificering van progenitor cel populaties binnen de ontwikkelingslanden cardiale halve maan, een belangrijke structuur gevormd tijdens de ontwikkeling van het hart. De aanpak is zodanig ontworpen dat zowel cel - en weefsel-niveau-informatie kan worden verkregen. Confocal microscopie en beeldverwerking kunt, dit protocol voor driedimensionale ruimtelijke wederopbouw van de cardiale crescent, waardoor de mogelijkheid om het analyseren van de lokalisatie en de organisatie van specifieke voorlopercellen populaties tijdens deze kritieke fase van de ontwikkeling van het hart. Nog belangrijker is, zorgt het gebruik van referentie antilichamen voor opeenvolgende maskeren van de cardiale halve maan en latere kwantitatieve metingen van gebieden binnen de halve maan. Dit protocol wordt niet alleen ingeschakeld met een grondig onderzoek van de vroege ontwikkeling van het hart, maar met aanpassingen gelden de meeste orgaansystemen in het gastrula vroege Somiet stadium muis embryonale.

De kwaliteit van de laatste gegevens en analyse hangt sterk (1) de integriteit en de morfologie van de ontleed embryo's, (2) het gebruik van hoge specificiteit antilichamen en (3) de juiste instelling van de imaging parameters. Beschadigde embryo's zal verwarren de oppervlakte generatie proces en een belemmering vormen voor kwantitatieve analyse.
