Neutrofiele extracellulaire vallen (netten), werden voor het eerst beschreven door Brinkmann et al. 1 ze zijn overwegend geproduceerd in reactie op de infectie (vooral voor bacteriën) en hebben een belangrijke rol in host defense1,2. Ze zijn ook beschreven moet worden uitgevoerd in reactie op niet-besmettelijke ziekte, met inbegrip van vasculitis en systemische lupus erythematosus (SLE); en naar de mitogeen phorbol 12-myrisate 13-acetaat (PMA)2,3. Het is onlangs erkend dat andere celtypes extracellulaire vallen, met inbegrip van de macrofagen, ook kunnen produceren. Macrophage extracellulaire vallen (METs) zijn nog niet een goed gedefinieerde entiteit in de literatuur4,5. We hebben onlangs gevestigde methoden voor het detecteren van de aanwezigheid van METs zowel in vitro als in vivo6,7. In dit artikel, zal de meting van METs met behulp van de confocal microscopie worden beschreven.
Zeer belangrijke eigenschappen van NETosis die zich van andere cellulaire routes (zoals apoptosis8 onderscheidt) zijn de extrusie van de chromatine in combinatie met: (1) Citrullinatie van histones (H3Cit)9, (2) mede uiting van de submodule proteasen10 , en (3) betrokkenheid van peptide arginine deiminase (PAD) 411,12. Macrofagen express ook H3Cit, submodule proteasen en PAD en deze functies kunnen worden gebruikt voor het definiëren van de aanwezigheid van METs.
METs hebben soms een bijzonder belangrijke rol in de longen, de macrofaag is de dominante cel aanwezig in de longblaasjes en de luchtwegen van de longen en heeft de eerste rol in de leiding van de cellulaire immuunrespons infectie/ontsteking. Bovendien, terwijl een groot deel van de Long is de lege ruimte (bijvoorbeeldbinnen de longblaasjes), kunnen de METs potentieel uit te breiden in de ruimte beschikbaar is, in tegenstelling tot solide organen.
De meest gebruikte methode om te definiëren van de aanwezigheid van netten is door confocale microscopie. Er is nog niet een duidelijk omschreven manier om te meten METs. De techniek voor de meting van de netten is aangepast voor het meten van de aanwezigheid van METs in dit protocol zijn. De belangrijkste vereisten voor deze methode zijn toegang tot confocale microscopie en passende denkbaar software voor analyse.

visualizing macrophage extracellular traps using confocal microscopy

Een primaire methode gebruikt om te definiëren van de aanwezigheid van neutrofiele extracellulaire vallen (netten) is confocale microscopie. Wij hebben gevestigde confocale microscopie methodes om te visualiseren macrofaag extracellulaire vallen (METs) bewerkt. Deze extracellulaire traps worden gedefinieerd door de aanwezigheid van extracellulaire chromatine met co uitdrukking van andere onderdelen zoals de submodule proteasen, citrullinated histonen en peptide arginase deiminase (PAD). De expressie van METs is over het algemeen gemeten na blootstelling aan een stimulans en vergeleken met un-gestimuleerd monsters. Monsters zijn ook opgenomen voor achtergrond en isotype controle. Cellen worden geanalyseerd met behulp van welomschreven afbeelding analysesoftware. Confocale microscopie kan worden gebruikt om te definiëren van de aanwezigheid van METs zowel in vitro als in vivo in longweefsel.

METs kunnen worden gevisualiseerd van BAL monsters, in longweefsel en dikker Long secties met 3D-beelden. Een voorbeeld van METs gevisualiseerd in een monster van de BAL is weergegeven in Figuur 1.
