最後の十年では、神経変性疾患、アルツハイマー病 (AD)、パーキンソン病 (PD) などの有病率は成長しています。これらの加齢に伴う疾患はこれらの患者の頭脳の線維性構造タンパク質凝集体の出現によって特徴付けられます。通常水溶性タンパク質を受けるまさに、集計プロセスは不十分な理解のままになります。蛋白質の集合は病気プロセスに限定されない代わりに、正常な老化プロセスの一部により異所萌出へを使用せずタンパク質凝集を調節する分子・細胞メカニズムの研究発見表現人間疾患関連タンパク質。ここで相補的なアプローチを通じてカエノラブディティス ・ エレガンスにおける固有の蛋白質凝集を調べるための方法論について述べる。まず、高齢の動物を取得する時代同期のc. の elegansの大量に成長する方法について説明、提案する高い不溶性-大きな凝集体を分離する生化学的なプロシージャ。ターゲット遺伝子ノックダウンと組み合わせて、促進またはいずれか包括的な分析を用いた定量的質量分析法によって年齢依存した蛋白質の集合を防ぐことの興味の遺伝子の役割を分析することが可能だか、抗体の候補者ベースの分析。これらの調査結果、集計しやすい蛋白質の蛍光タグを表現するトランスジェニック動物を体内分析によるに確認します。これらのメソッドは、ある特定の蛋白質が加齢とともに集計と最終的にどのようにこれらの蛋白質を完全に機能しやすい理由を明確に役立つはずです。
