Avec le développement rapide des techniques analytiques modernes, des centaines de composés organiques volatils (COV) ont été identifiées dans air expiré humaine1. Ces COV résultant principalement de l’air alvéolaire (~ 350 mL pour un adulte en bonne santé) et l’air de l’espace mort anatomique (~ 150 mL)2, qui sont touchés par l’organisme métabolisme3,4,5,6,7 ,8 et9de la pollution environnementale, respectivement. Ainsi, si identifiés, ces COV est prometteurs à être utilisés comme biomarqueurs pour le diagnostic de la maladie et l’exposition environnementale d’une manière non invasive.
Bien que la spectrométrie de masse de chromatographie en phase gazeuse (GC-MS) est la technique la plus utilisée pour l’analyse qualitative et quantitative des exhalé COV2, direct MS techniques, qui ont été développées pour l’analyse d’haleine en temps réel, ont les avantages de grand temps de résolution et préparation préalable de l’échantillon simple. Direct MS techniques, comme la réaction de transfert de proton de MS (PTR-MS)10, sélectionné tube de débit ion MS (SIFT-MS)11, secondaire electrospray ionisation MS (SESI-MS)12,13 (également appelés electrospray extractive ionisation, MS, MS-EESI14,,15), trace des gaz atmosphériques analyzer (TAGA)16 et plasma ionisation MS (PI-MS)17 ont été étudiées dans ces dernières années.
Parmi toutes les techniques MS directs, SESI est bien connue comme une ionisation douce universelle technique19,20,21; et la source est facile à être personnalisés et couplé aux différents types de spectromètres de masse, par exemple, temps de vol spectromètre de masse8,15, piège à ions de masse spectromètre14 et Orbitrap valant spectromètre de masse12 ,,18. Jusqu'à présent, SESI-MS a été utilisé avec succès dans le diagnostic des maladies respiratoires22, rythme circadien3,6,23, pharmacocinétique7,8, de jaugeage et révélant des voies métaboliques4, etc.. Plus récemment, une source commerciale de SESI est devenu disponible.
Dans cette étude, une source d’ionisation facile et compact SPECTROMETRIE secondaire (Sec-nanoESI) a été mises en place et couplée à un spectromètre de masse à haute résolution. Mesures en temps réel des COV expiré en haleine ont été présentés.

real-time breath analysis by using secondary nanoelectrospray ionization coupled to high resolution mass spectrometry

Exhalé de composés organiques volatils (COV) ont suscité un intérêt considérable, puisqu’ils peuvent servir de biomarqueurs pour le diagnostic de la maladie et l’exposition environnementale d’une manière non invasive. Dans ce travail, nous présentons un protocole afin de caractériser les COV expirés en temps réel en utilisant l’ionisation de SPECTROMETRIE secondaire couplée à la spectrométrie de masse haute résolution (Sec-nanoESI-HRMS). La source s-nanoESI maison est facilement mises en place basée sur une source commerciale nanoESI. Des centaines de pics ont été observées dans les spectres de masse soustrait au fond de l’air expiré, et les valeurs d’exactitude de masse sont -4,0-13,5 ppm et -20,3-1,3 ppm dans les modes de détection des ions positifs et négatifs, respectivement. Les sommets ont été assignés avec composition élémentaire exacte selon le modèle isotopique et de la masse exacte. Moins de 30 s est utilisé pour la mesure d’une expiration, et il faut environ 7 min pour les six mesures répétées.
