Con rápido desarrollo de técnicas analíticas modernas, se han identificado cientos de compuestos orgánicos volátiles (COV) en aire espirado humana1. Estos VOCs resultan sobre todo de aire alveolar (~ 350 mL en un adulto sano) y el espacio muerto anatómico del aire (~ 150 mL)2, que son afectadas por cuerpo metabolismo3,4,5,6,7 ,8 y9de la contaminación ambiental, respectivamente. Como resultado, si, estos VOCs son prometedores para utilizarse como biomarcadores para diagnóstico de enfermedades y la exposición ambiental de una manera no invasiva.
Cromatografía de gases espectrometría de masas (GC-MS) es la técnica más ampliamente utilizada para el análisis cualitativo y cuantitativo de exhalado COV2, técnicas directas de MS, que se han desarrollado para el análisis en tiempo real de la respiración, tienen las ventajas de hora de resolución y preparación previa de muestra simple. Técnicas directas de MS, como la reacción de transferencia de protones MS (PTR-MS)10, seleccionan tubo de flujo de iones MS (SIFT-MS)11, secundaria electrospray ionización MS (SESI-MS)12,13 (también nombrado como extractiva electrospray ionización, MS, MS EESI14,15), trace gas atmosférico analizador (TAGA)16 y plasma ionización MS (PI-EM)17 han sido investigados en años recientes.
Entre todas las técnicas de MS directas, SESI es conocido como una ionización suave universal técnica19,20,21; y la fuente es fácil de personalizar y junto a diferentes tipos de espectrómetros de masas, por ejemplo, el tiempo de vuelo espectrómetro de masas8,15, trampa de iones masa espectrómetro14 y orbitrap espectrómetro de masas12 ,18. Exhalado compuestos orgánicos volátiles (COV) han despertado un gran interés, ya que pueden servir como biomarcadores para el diagnóstico de la enfermedad y la exposición ambiental de una manera no invasiva. En este trabajo, presentamos un protocolo para caracterizar los COVs exhalados en tiempo real mediante el uso de ionización secundaria nanoelectrospray acoplada a espectrometría de masas de alta resolución (s-nanoESI-HRMS). La fuente Sec nanoESI casera fácilmente nace basada en una fuente de nanoESI comercial. Se observaron cientos de picos en los espectros de masa fondo resta de aire espirado, y los valores de exactitud total son-4.0-13,5 ppm y-20.3-1,3 ppm en los modos de detección de iones positivos y negativos, respectivamente.
