De bepaling van de osteoclastogenic is de wijdst gebruikte methode cultuur osteoclasten in vitro12,13te isoleren. Terwijl verscheidene RANKL gebaseerde osteoclast inducties beschreven13,14,15 zijn, wordt in de huidige studie een protocol met enkele aanpassingen op basis van de bovenstaande methoden beschreven.

Bot is een orgaan steeds veranderende en is gerenoveerd door botcellen en osteoclasten gedurende het hele leven. Osteoclasten zijn verantwoordelijk voor gemineraliseerde matrix resorptie en botcellen synthetiseren en afscheiden van nieuw bot matrices1. Het evenwicht tussen resorptie van het been en been vorming is van cruciaal belang voor de gezondheid van de botten met inbegrip van onderhoud van bot massa en respons op stimulatie en schade. Als dit evenwicht is verstoord, kan een reeks van metabole ziekten bot optreden, met inbegrip van osteoporose en parodontale ziekten. In deze ziekten overschrijdt massale botverlies als gevolg van osteoclastic bot resorptie het bot vormen capaciteit van botcellen2,3. Het is dus, teneinde farmaceutische doelstellingen voor de behandeling van skeletstoornissen zoals osteoporose, kritiek te begrijpen van de generatie en de biologie van osteoclasten4.
Osteoclasten zijn unieke giant multinucleaire cellen zich op of in de buurt van het oppervlak van bot en behoren tot de familie van monocyt/macrofaag1. Ibbotson K. J. et al. een methode voor het genereren van osteoclast-achtige cellen in vitro met medium met 1,25-dihydroxy-vitamine D35gemeld. De identificatie van macrofaag-kolonie stimulerende factor (M-CSF) en receptor activator voor nuclear factor-Kappa B ligand (RANKL) als essentiële factoren van osteoclast formatie is sterk toegenomen de efficiëntie van de osteoclastogenesis in vitro 1 , 6 , 7. de mogelijkheid om cultuur osteoclasten in vitro verbeterd ons begrip van de generatie en regulering van de osteoclasten.
Het zoogdier/mechanistische doel van rapamycin (mTOR) functies in twee structureel en functioneel verschillende complexen, namelijk mTORC1 en mTORC28,9. De twee multi eiwitcomplexen zijn verschillend van elkaar als gevolg van hun verschillende componenten en de stroomafwaartse substraten. mTORC1 bevat de unieke regelgevende-geassocieerde proteïne van mTOR (Raptor), terwijl mTORC2 de rapamycin-ongevoelig metgezel van mTOR (Rictor)9 bevat. mTORC1 kan integreren en zenden belangrijke signalen voor het regelen van de celgroei, proliferatie en differentiatie. Onlangs, we laten zien dat mTORC1 speelt een sleutelrol in het netwerk van katabole bot resorptie door schrapping van de Raptor naar de inactivering van mTORC1 in de osteoclasten10. Echter de onderliggende mechanismen nader moeten worden onderzocht. In de huidige studie, werd een RANKL gebaseerde osteoclastogenic-methode gebruikt voor het genereren van osteoclasten uit beenmerg-afgeleide macrofagen (BMMs) van wild-type (WT) en RapCtsk muizen, en te bestuderen van de invloed van mTORC1 inactivatie op osteoclast vorming.

Osteoclasten zijn unieke bot-resorbeerbare cellen die van de bloedlijn monocyt/macrophage van beenmerg onderscheiden. Dysfunctie van osteoclasten kan resulteren in een reeks van bot metabole ziekten, met inbegrip van osteoporose. Ontwikkeling van farmaceutische doelstellingen voor de preventie van pathologische massa botverlies, moeten de mechanismen waarmee osteoclasten van precursoren onderscheiden worden begrepen. De mogelijkheid om te isoleren en een groot aantal osteoclasten in vitro cultuur is van cruciaal belang om te bepalen van de rol van specifieke genen bij osteoclast differentiatie. Inactivering van het zoogdier/mechanistische doelwit van rapamycin complexe 1 (TORC1) in osteoclasten kan verminderen osteoclast nummer en verhoging van de botmassa; echter de onderliggende mechanismen nader moeten worden onderzocht. In de huidige studie, wordt een RANKL gebaseerde protocol cultuur osteoclasten uit beenmerg van de muis te isoleren en te bestuderen of de invloed van mTORC1 inactivatie op osteoclast vorming beschreven. Dit protocol met succes geleid tot een groot aantal reus osteoclasten, meestal binnen een week. Schrapping van de Raptor verminderde osteoclast vorming en daalde de activiteit van secretoire tartraat-resistente zure fosfatase, die aangeeft dat mTORC1 is van cruciaal belang voor osteoclast vorming.

Met behulp van dit protocol, werden een groot aantal reus osteoclasten gezien op dag 6; Als giant osteoclasten niet gezien zijn, een dag van osteoclast differentiatie mogelijk nodig (Figuur 1).
