OS est un organe en constante évolution et est remodelée par les ostéoblastes et les ostéoclastes tout au long de la vie. Les ostéoclastes sont responsables de la résorption de la matrice minéralisée et ostéoblastes synthétisent et sécrètent des nouveaux OS matrices1. L’équilibre entre la résorption et formation osseuse est cruciale pour la santé osseuse, y compris l’entretien des os masse et réponse à la stimulation et de blessures. Si cet équilibre est rompu, une série de maladies métaboliques des os peut se produire, y compris l’ostéoporose et les maladies parodontales. Dans ces maladies, la perte de masse osseuse résultant de la résorption osseuse ostéoclastique dépasse les os formant la capacité des ostéoblastes2,3. Ainsi, afin d’élaborer des cibles pharmaceutiques pour traiter des troubles squelettiques telles que l’ostéoporose, il est essentiel de comprendre la génération et la biologie des ostéoclastes4.
Les ostéoclastes sont des cellules multinucléées géants uniques situés à ou près de la surface osseuse et appartiennent à la famille de monocytes/macrophages1. Ibbotson K. J. et al. a signalé une méthode pour générer des cellules semblables à des ostéoclastes in vitro avec un milieu contenant 1, 25-dihydroxy-vitamine D35. L’identification du facteur stimulant de macrophage-colony (M-CSF) et activateur du récepteur pour le ligand de facteur nucléaire-κ B (RANKL) comme facteurs essentiels de la formation des ostéoclastes a considérablement accru l’efficacité des osteoclastogenesis in vitro 1 , 6 , 7. la capacité de la culture les ostéoclastes in vitro a amélioré notre compréhension de la génération et la réglementation des ostéoclastes.
La cible mammifère/mécaniste de la rapamycine (mTOR) fonctions dans deux complexes structurellement et fonctionnellement distinctes, à savoir mTORC1 et mTORC28,9. Les deux complexes de protéines multiples sont distincts les uns des autres en raison de leurs différents composants et des substrats en aval. mTORC1 contient la protéine associée à la réglementation unique de mTOR (rapace), tandis que mTORC2 contient la rapamycine insensible compagnon de mTOR (Rictor)9. mTORC1 peut intégrer et transmettre des signaux importants pour réguler la croissance cellulaire, la prolifération et la différenciation. Récemment, nous avons démontré que mTORC1 joue un rôle clé dans le réseau de la résorption osseuse catabolique par suppression du Raptor pour inactiver les mTORC1 dans les ostéoclastes10. Cependant, les mécanismes sous-jacents nécessitent une étude plus approfondie. Dans la présente étude, une méthode axée sur le RANKL osteoclastogenic a été utilisée pour générer des ostéoclastes de macrophages dérivés de la moelle osseuse (BMMs) de type sauvage (WT) et de la souris Ctsk de Rap et d’étudier l’influence de l’inactivation de mTORC1 sur les ostéoclastes formation.

Les ostéoclastes sont unique-résorption osseuse des cellules qui se différencient de la lignée des monocytes/macrophages de la moelle osseuse. Une série de maladies métaboliques des os, dont l’ostéoporose peut entraîner un dysfonctionnement des ostéoclastes. Pour développer des cibles pharmaceutiques pour la prévention de la perte de masse osseuse pathologique, faut comprendre les mécanismes par lesquels les ostéoclastes différencient des précurseurs. La capacité d’isoler et de la culture d’un grand nombre d’ostéoclastes in vitro est cruciale afin de déterminer le rôle des gènes spécifiques dans la différenciation des ostéoclastes. Inactivation de la cible mammifère/mécaniste de la rapamycine complexe 1 (TORC1) dans les ostéoclastes peut diminuer le nombre d’ostéoclastes et augmenter la masse osseuse ; Cependant, les mécanismes sous-jacents nécessitent une étude plus approfondie. Dans la présente étude, on décrit un protocole RANKL d’isoler et de culture d’ostéoclastes de moelle osseuse de souris et d’étudier l’influence de l’inactivation de mTORC1 sur la formation des ostéoclastes. Ce protocole a permis avec succès dans un grand nombre des ostéoclastes géants, en général sous huitaine. Suppression du Raptor avec facultés affaiblies de la formation des ostéoclastes et diminue l’activité de sécrétion phosphatases acides tartrate-résistantes, qui indique que mTORC1 est essentielle pour la formation des ostéoclastes.
