Knochen ist ein ständig wechselnden Organ und wird von Osteoblasten und Osteoklasten zeitlebens umgebaut. Osteoklasten sind verantwortlich für mineralisierten Matrix Resorption und Osteoblasten zu synthetisieren und sezernieren neue Knochen Matrizen1. Die Balance zwischen Knochenabbau und Knochenaufbau ist entscheidend für die Knochengesundheit, einschließlich der Wartung der Knochen Masse und Reaktion auf Stimulation und Verletzungen. Wenn dieses Gleichgewicht gestört ist, kann eine Reihe von metabolischen Knochenerkrankungen wie Osteoporose und parodontale Erkrankungen auftreten. Bei diesen Erkrankungen übersteigt Knochen Masse Verlust infolge osteoclastic Knochenabbau Knochen bilden Kapazität von Osteoblasten2,3. So um pharmazeutische Ziele zur Behandlung von Skeletterkrankungen wie Osteoporose zu entwickeln, ist es wichtig zu verstehen, die Erzeugung und die Biologie der Osteoklasten4.
Osteoklasten sind einzigartige mehrkernigen Riesenzellen befindet sich an oder nahe der Knochenoberfläche und gehören zu der Familie Monocyte/Makrophagen-1. Ibbotson K. J. Et al. eine Methode zur Erzeugung Osteoklasten-wie Zellen in Vitro mit 1,25-Dihydroxy-Vitamin D35-haltigem Medium berichtet. Die Identifizierung von Makrophagen-Kolonie-stimulierende Faktor (M-CSF) und Rezeptor Aktivator für nukleare Faktor κ B Ligand (RANKL) als wesentliche Faktoren der Osteoklasten Bildung hat dramatisch zugenommen, die Effizienz der Osteoclastogenesis in vitro 1 , 6 , 7. die Möglichkeit, Kultur Osteoklasten in Vitro hat unser Verständnis der Generation und Regulierung der Osteoklasten verbessert.
Das Säugetier/mechanistischen Ziel von Rapamycin (mTOR) Funktionen in zwei strukturell und funktionell unterschiedliche komplexe, nämlich mTORC1 und mTORC28,9. Die zwei Multi-Protein-komplexe sind aufgrund ihrer unterschiedlichen Komponenten und nachgelagerten Substrate voneinander. mTORC1 enthält das einzigartige regulatorischen-assoziierten Protein mTOR (Raptor), während mTORC2 Rapamycin-unempfindliche Begleiter von mTOR (Rictor)9enthält. mTORC1 können integrieren und übertragen wichtige Signale, Zellwachstum, Proliferation und Differenzierung zu regulieren. Vor kurzem haben wir gezeigt, dass diese mTORC1 spielt eine Schlüsselrolle im Netzwerk der katabolen Knochenabbau durch Löschen des Raptor , mTORC1 in Osteoklasten10zu inaktivieren. Jedoch bedürfen die zugrunde liegenden Mechanismen weiterer Untersuchungen. In der vorliegenden Studie wurde eine Osteoclastogenic RANKL-basierte Methode Osteoklasten aus dem Knochenmark stammenden Makrophagen (BMMs) der Wildtyp (WT) und RapCtsk Mäuse zu generieren und Untersuchung des Einflusses von mTORC1 Inaktivierung auf Osteoklasten verwendet. Osteoklasten sind einzigartige Knochen-resorbierbarem Zellen, die von der Monocyte/Makrophagen-Linie des Knochenmarks zu unterscheiden. Dysfunktion von Osteoklasten kann eine Reihe von metabolischen Knochenerkrankungen wie Osteoporose führen. Inaktivierung von Säugetieren/mechanistischen Ziel von Rapamycin Komplex 1 (TORC1) in Osteoklasten kann Osteoklasten Anzahl verringern und Knochenmasse zu erhöhen; jedoch bedürfen die zugrunde liegenden Mechanismen weiterer Untersuchungen. In der vorliegenden Studie wird ein RANKL-basiertes Protokoll zu isolieren und Kultur Osteoklasten aus Maus Knochenmark und Untersuchung des Einflusses von mTORC1 Inaktivierung auf Osteoklasten Formation beschrieben. Dieses Protokoll führte erfolgreich eine große Anzahl von riesigen Osteoklasten in der Regel innerhalb einer Woche. Löschen von Raptor beeinträchtigt Osteoklasten Bildung und nahm die Aktivität des sekretorischen Tartrat-resistente saure Phosphatase, anzeigend, dass mTORC1 für Osteoklasten Bildung entscheidend.

Mit Hilfe dieses Protokolls, wurden eine große Anzahl von riesigen Osteoklasten am 6.
