O ensaio de osteoclastogenic é o método mais utilizado para isolar e cultura osteoclastos em vitro12,13. Enquanto várias induções baseadas em RANKL osteoclast têm sido descritos13,14,15, o presente estudo descreveu um protocolo com algumas modificações com base em métodos anteriores.

Osso é um órgão em constante mudança e é remodelado por osteoblastos e osteoclastos ao longo da vida. Osteoclastos são responsáveis pela reabsorção de matriz mineralizada e osteoblastos sintetizam e segregam o novo osso matrizes1. O equilíbrio entre a reabsorção óssea e formação óssea é crucial para a saúde óssea, incluindo a manutenção do osso em massa e resposta à estimulação e lesão. Se esse equilíbrio é perturbado, pode ocorrer uma série de doenças metabólicas do osso, incluindo osteoporose e doenças periodontais. Estas doenças, perda de massa óssea resultante da reabsorção óssea osteoclástica excede o osso formando a capacidade de osteoblastos2,3. Assim, a fim de desenvolver metas farmacêuticas para tratar distúrbios esqueléticos, tais como osteoporose, é fundamental para compreender a geração e a biologia dos osteoclastos4.
Osteoclastos são únicas células multinucleadas gigantes localizadas em ou perto da superfície do osso e pertencem à família de monócitos/macrófagos1. Ibbotson K. J. et al. relatou um método para gerar células osteoclasto em vitro com meio contendo 1,25-diidroxi-vitamina D35. A identificação do fator estimulante de colônia de macrófagos (M-CSF) e ativador do receptor para o ligante de B fator nuclear-κ (RANKL) como fatores essenciais da formação do osteoclast aumentou drasticamente a eficiência do osteoclastogenesis em vitro 1 , 6 , 7. a capacidade de cultura osteoclastos em vitro melhorou a nossa compreensão da geração e regulação dos osteoclastos.
Mamíferos/mecanicista alvo de rapamicina (mTOR) funções em dois complexos estruturalmente e funcionalmente distintos, ou seja, mTORC1 e mTORC28,9. Os dois complexos de proteínas multi são distintos uns dos outros devido a seus diferentes componentes e substratos a jusante. mTORC1 contém a proteína reguladora associada exclusiva de mTOR (Raptor), enquanto mTORC2 contém o companheiro rapamicina diferenciação da mTOR (Rictor)9. mTORC1 pode integrar e transmitir sinais importantes que regulam crescimento celular, proliferação e diferenciação. Recentemente, temos demonstrado que mTORC1 desempenha um papel fundamental na rede de reabsorção óssea catabólica pela exclusão de Raptor para inactivar mTORC1 em osteoclastos10. No entanto, os mecanismos subjacentes exigem um estudo mais aprofundado. No presente estudo, um método baseado em RANKL osteoclastogenic foi usado para gerar os osteoclastos de macrófagos derivados da medula óssea (BMMs) do selvagem-tipo (WT) e RapCtsk ratos e estudar a influência de inativação de mTORC1 no osteoclasto formação.

Osteoclastos são único osso-resorbing células que diferenciam-se da linhagem de monócitos/macrófagos da medula óssea. Disfunção dos osteoclastos pode resultar em uma série de doenças metabólicas ósseas, incluindo osteoporose. Para desenvolver metas farmacêuticas para a prevenção da perda de massa óssea patológica, devem-se compreender os mecanismos pelos quais osteoclastos diferenciarem dos precursores. A capacidade de isolar e cultura de um grande número de osteoclastos em vitro é fundamental para determinar o papel de genes específicos na diferenciação do osteoclast. Inactivação do alvo da rapamicina complexo 1 (TORC1) em osteoclastos mamíferos/mecanicista pode diminuir o número de osteoclasto e aumento da massa óssea; no entanto, os mecanismos subjacentes exigem um estudo mais aprofundado. No presente estudo, é descrito um protocolo baseado em RANKL isolar e cultura osteoclastos da medula óssea de rato e estudar a influência de inativação de mTORC1 na formação do osteoclast. Este protocolo com sucesso resultou em um grande número de osteoclastos gigantes, normalmente dentro de uma semana. Exclusão de Raptor deficiente formação osteoclasto e diminuiu a atividade secretora fosfatase ácida tartarato-resistente, indicando que mTORC1 é fundamental para a formação do osteoclast.
