Denne artikel viser en murine model for at studere udviklingen af myointimal hyperplasi og giver mulighed for udforskning af de underliggende patologiske processer og afprøvning af nye lægemidler eller terapeutiske muligheder.
Det mest afgørende skridt i denne protokol er denudation af aorta. Særlig omsorg bør lægges under dette trin, da overdreven denudation vil føre til aneurisme dannelse og model fiasko. På den anden side, hvis denudation udføres tilstrækkeligt, vil for lidt myoin...

Arteriel stenose i koronar og perifere arterier har en stor effekt på sygelighed og dødelighed af patienter1. En underliggende patologiske mekanisme er myointima hyperplasi (MH), som er karakteriseret ved øget spredning, migration og syntese af ekstracellulære matrix proteiner fra vaskulære glatte muskelceller celler (SMC)2. SMC er beliggende i media lag af fartøjet og overfører ved stimulation til overfladen af lumen. Stimulerende signaler inkluderer vækstfaktorer cytokiner, celle-celle kontakt, lipider, ekstracellulære matrix komponenter og mekaniske shear og strække styrker3,4,5,6. Skader af karvæggen, patologiske eller iatrogen, forårsage endotel celle og glatte muskulatur celleskader og stimulere inflammatoriske reaktioner, og således føre til MH7.
Forskellige dyremodeller er i øjeblikket tilgængelig at studere arteriel skade og myointima hyperplasi. Store dyr som grise eller hunde har fordelen, at sharing en lignende arterie og koronar anatomi med mennesker og er specielt velegnet til studier undersøger angioplastik teknikker, procedure og enheder8. Gris modeller har dog Ulempen ved højere thrombogenicity9,10, mens hunde kun har en mild reaktion fartøj skade11. Desuden, kræver alle store dyremodeller særlige boliger, udstyr og personale, som er forbundet med høje omkostninger og er ikke altid tilgængelige på en institution. Lille dyremodeller omfatter rotter og mus. Sammenlignet med rotter, har mus fordelene ved lavere omkostninger og eksistensen af en bred vifte af knock ud modeller. Den model, der er beskrevet i denne video kan kombineres med ApoE-/-mus fodret med en vestlig kost at nøje efterligne de kliniske omgivelser for ballonudvidelse i aterosklerotisk fartøjer12. Tidligere modeller induceret vaskulære skade via wire skade13, flydende udtørring14, foråret15eller manchet skade16. Da arten af skaden vil høj grad påvirke udviklingen og forfatning af MH, er ved hjælp af en ballonkateter for at fremkalde fartøj skade den bedste måde at efterligne de kliniske omgivelser.
I denne artikel beskriver vi en ny metode for at fremkalde MH med en ballonkateter i mus. Brug af et ballonkateter (1,2 mm x 6 mm) med en RX-Port (figur 1A) tillader skrabning af det intima lag og samtidig induktion af en overdistension af fartøjet. Begge disse faktorer er vigtige udløsere for udviklingen af MH. Observationstidspunkt for denne model er 28 dage17.

Anvendelsen af dyremodeller er afgørende for en bedre forståelse af MH, en væsentlig årsag til arteriel stenose. I denne artikel viser vi en murine ballon denudation model, der kan sammenlignes med etablerede fartøj skade modeller i store dyr. Aorta denudation model med ballon katetre efterligner de kliniske omgivelser og fører til sammenlignelige pathobiological og fysiologiske ændringer. Kort, efter at have udført et vandret snit i aorta abdominalis, en ballonkateter vil være indsat i fartøjet, oppustet og indført retrogradely. Oppustning af ballonen vil føre til intima skade og overdistension af fartøjet. Efter fjernelse af kateteret, vil aorta indsnit blive lukket med enkelt masker.
