Cet article illustre un modèle murin pour étudier le développement de l’hyperplasie myointimal et permet l’exploration des processus pathologiques sous-jacent et les essais de nouveaux médicaments ou les options thérapeutiques.

La sténose artérielle dans les artères coronaires et périphériques a un effet important sur la morbidité et la mortalité des patients1. Un mécanisme pathologique sous-jacent est l’hyperplasie myointima (MH), qui se caractérise par une augmentation de la prolifération, la migration et la synthèse des protéines de la matrice extracellulaire du muscle lisse vasculaire des cellules (SMC)2. SMC se trouvent dans la couche de support du navire et de migrer lors de la stimulation à la surface de la lumière. Signaux stimulants incluent les facteurs de croissance, cytokines, contact cellule-cellule, lipides, les composants de la matrice extracellulaire et cisaillement mécanique et étirement les forces3,4,5,6. Lésions de la paroi des vaisseaux, pathologique ou iatrogène, causent des cellules endothéliales et les dommages aux cellules de muscle lisse et stimulent les réactions inflammatoires et donc conduisent à MH7.
Différents modèles animaux sont actuellement disponibles pour étudier les lésions artérielles et hyperplasie myointima. Grands animaux comme les porcs ou les chiens ont l’avantage d’une artère similaire et l’anatomie coronaire de partage avec les humains et sont particulièrement adaptées aux études portant sur l’angioplastie techniques, procédures et dispositifs8. Cependant, modèles de cochon ont l’inconvénient de supérieur thrombogénicité9,10, tandis que les chiens n’ont une réponse douce à navire blessures11. En outre, tous les grands modèles animaux nécessitent logement spécifique, l’équipement et le personnel, qui est lié à des coûts élevés et n’est pas toujours disponibles dans une institution. Petits modèles animaux comprennent les rats et les souris. Comparativement à des rats, des souris ont les avantages d’un coût moindre et l’existence d’une variété d’assommer de modèles. Le modèle décrit dans cette vidéo peut être associé d’ApoE-/-souris nourries avec un régime occidental pour imiter étroitement le contexte clinique d’angioplastie dans navires athéroscléreuse12. Les modèles précédents induit des lésions vasculaires via fil blessures13, dessiccation fluide14, printemps15ou des rotateurs blessures16. Étant donné la nature de la lésion affectera grandement le développement et la constitution de MH, à l’aide d’un cathéter à ballonnet pour induire la lésion en est le meilleur moyen d’imiter le contexte clinique.
Dans cet article, nous décrivons une nouvelle méthode pour induire des MH avec un cathéter à ballonnet chez la souris. L’utilisation d’un cathéter à ballonnet (1,2 mm x 6 mm) avec un RX-Port (Figure 1 a) permet le grattage de la couche intimale et, en même temps, l’induction d’une surdistension du navire. Ces deux facteurs sont des déclencheurs importants pour le développement de MH. L’heure de l’observation pour ce modèle est de 28 jours,17.

L’utilisation de modèles animaux est essentielle pour une meilleure compréhension de la MH, une des principales causes de la sténose artérielle. Dans cet article, nous démontrons un modèle murin de ballon dénudation, qui est comparable aux modèles de lésion du navire établies dans les grands animaux. Le modèle de dénudation aorte avec des cathéters à ballonnet imite le contexte clinique et conduit à des changements physiologiques et pathobiological comparables. En bref, après avoir effectué une incision horizontale dans l' aorte abdominalis, un cathéter à ballonnet sera inséré dans le vaisseau, gonflé et introduit marqués. Gonflage du ballonnet conduira à des blessures de l’intima et une surdistension du navire. Après avoir retiré le cathéter, l’incision aortique sera fermée avec des points de suture unique. Le modèle présenté dans cet article est reproductible, facile à réaliser et peut être établie de manière rapide et fiable. Il est particulièrement approprié pour l’évaluation de coûteux agents thérapeutiques expérimentales, qui peuvent être appliqués de façon économique. Dénudation du ballon est un modèle approprié pour étudier le développement du MH chez la souris.
