Arterielle Stenose in den koronaren und peripheren Arterien hat einen großen Effekt auf die Morbidität und Mortalität von Patienten1. Eine zugrunde liegende Pathomechanismus ist Myointima Hyperplasie (MH), charakterisiert durch erhöhte Proliferation, Migration und Synthese von Proteinen der extrazellulären Matrix von vaskulären glatten Muskelzellen Zellen (SMC)2. SMC befinden sich in der Medienschicht des Schiffes und nach Stimulation an die Oberfläche des Gefäßlumens zu migrieren. Stimulierende Signale sind Wachstumsfaktoren, Zytokine, Zell-Zell-Kontakt, Lipide, Komponenten der extrazellulären Matrix und mechanische Scherkräfte und Strecken Kräfte3,4,5,6. Verletzungen der Gefäßwand, pathologische oder iatrogene, Endothelzellen und glatten Muskelzellen Zellschäden verursachen Entzündungsreaktionen zu stimulieren und somit zu MH7führen.
Verschiedene Tiermodellen sind derzeit für arterielle Verletzungen und Myointima Hyperplasie zu studieren. Große Tiere wie Schweine oder Hunde haben den Vorteil, eine ähnliche Arterie und koronare Anatomie mit Menschen zu teilen und eignen sich besonders für Studien zur Untersuchung der Angioplastie Techniken, Verfahren und Geräte8. Schwein-Modelle weisen jedoch den Nachteil der höheren Thrombogenität9,10, während Hunde nur eine milde Reaktion auf Schiff Verletzungen11haben. Darüber hinaus müssen alle großen Tiermodellen Spezialgehäuse, Ausrüstung und Personal, die sind mit hohen Kosten verbunden und stehen nicht immer an einer Hochschule. Kleiner Tiermodelle sind Ratten und Mäuse. Haben im Vergleich zu Ratten, Mäuse, die Vorteile der geringeren Kosten und die Existenz einer Vielzahl von k.o.-Modelle. Das Modell in diesem Video beschrieben kombinierbar mit ApoE-/-Mäuse mit einer westlichen Diät gefüttert, genau die klinische Einstellung der Angioplastie in atherosklerotischen Schiffe12imitieren. Frühere Modelle induziert Gefäßverletzung per Draht Verletzungen13, Fluid Austrocknung14, Frühjahr15oder Manschette Verletzungen16. Da die Art der Verletzung erheblich die Entwicklung und die Verfassung der MH auswirken wird, ist mit einem Ballonkatheter induzieren Schiff Verletzungen der beste Weg, um der klinischen Einstellung zu imitieren.
In diesem Artikel beschreiben wir eine neuartige Methode zur MH mit einem Ballonkatheter bei Mäusen zu induzieren. Die Verwendung eines Ballon-Katheters (1,2 x 6 mm) mit einem RX-Anschluss (Abbildung 1A) ermöglicht das Kratzen der Intima Schicht und gleichzeitig die Induktion von einem Overdistension des Schiffes. Beide Faktoren sind wichtige Auslöser für die Entwicklung von MH. Die Beobachtungszeit für dieses Modell beträgt 28 Tage17.

Ballon Denudation ist ein geeignetes Modell für die Entwicklung von MH bei Mäusen zu studieren. Tiere gut von der Operation erholen und zeigen eine ausgezeichnete Kondition nach der Operation.
